摘要
目的研究瑞舒伐他汀和阿托伐他汀降脂疗效与ABCB1 G2677T基因多态性的相关性。方法采用实用性随机对照试验,将收集符合标准的483例ABCB1 G2677T基因检测患者分为瑞舒伐他汀组和阿托伐他汀组,分析瑞舒伐他汀和阿托伐他汀降脂疗效与ABCB1基因多态性之间的相关性。结果 483例患者的ABCB1G2677T基因型分布符合Hardy-Weinberg遗传平衡(P> 0.05),ABCB1 G2677T最小等位基因频率(MAF)为48.24%。服药前,2组患者血脂水平比较差异无统计学意义(P> 0.05)。服药4周后,血脂变化率瑞舒伐他汀组与阿托伐他汀组之间差异无统计学意义(P> 0.05);GG型、GT型患者瑞舒伐他汀组与阿托伐他汀组血脂变化率差异也无统计学意义(P> 0.05)。在TC、LDL脂质水平降幅中,TT型患者瑞舒伐他汀组与阿托伐他汀组比较差异有统计学意义(P <0.05),且瑞舒伐他汀组降幅高于阿托伐他汀组;在TG、HDL脂质水平降幅2组比较差异无统计学意义(P> 0.05)。结论瑞舒伐他汀和阿托伐他汀降脂疗效与ABCB1 G2677T基因多态性有相关性,ABCB1 G2677T携带TT等位基因患者选用瑞舒伐他汀降脂效果优于阿托伐他汀。
AIM To investigate the relationship between the effect of rosuvastatin and atorvastatin on reducing blood lipid and ABCB1 G2677 T gene polymorphism.METHODS A total of 483 patients with ABCB1 G2677 T gene by fluorescence staining in situ hybridization were divided into rosuvastatin group and atorvastation group by randomized controlled trial,and the association between the effect of rosuvastatin and atorvastatin on lipid-lowering and ABCB1 gene polymorphism was analyzed.RESULTS The distribution of ABCB1 G2677 T genotype in 483 patients was consistent with Hardy-Weinberg genetic balance(P>0.05),and the minor allele frequency(MAF)of ABCB1 G2677 T was 48.24%.There was no significant difference in blood lipid between rosuvastatin group and atorvastatin group(P>0.05).There was no significant difference in the change rate of blood lipid between rosuvastatin group and atorvastatin group after 4 weeks of administration(P>0.05).There was no significant difference in the change rate of blood lipid in GT genotype and GG genotype patients between rosuvastatin group and atorvastatin group(P>0.05).In the decrease of TC and LDL lipid levels,there was a significant difference in TT genotype patients between rosuvastatin group and atorvastatin group(P<0.05),and the range of reducing blood lipid in rosuvastatin group was higher than that in atorvastatin group.There was no significant difference in TG and HDL lipid levels between rosuvastatin group and atorvastatin group(P>0.05).CONCLUSION There is the correlation between the effect of rosuvastatin and atorvastatin on reducing blood lipid and ABCB1 G2677 T gene polymorphism.Compared with atorvastatin,rosuvastatin is more effective in reducing blood lipid in TT genotype patients.
