摘要
目的:探讨肺复康方对肺癌癌因性疲乏裸鼠骨骼肌的影响。方法:将25只裸鼠随机分为空白对照组、模型组、肺复康方低、中、高剂量组,每组5只。除空白对照组外,其余4组裸鼠均皮下注射Lewis肺癌细胞,建立癌因性疲乏裸鼠模型,利用鼠尾悬挂实验进行疲乏测定。3周后进行药物干预,每周5天,同时根据鼠尾悬挂实验(2次/周)判断其疲乏缓解情况。6周后药物干预结束,取裸鼠骨骼肌进行分裂蛋白(Drp1)、融合蛋白(Mfn1)、ATP、丙二醛(MDA)以及谷胱甘肽(GSH)的检测。结果:药物干预前:与空白对照组相比,模型组及剂量组失望时间均延长。药物干预后:肺复康方能够下调肺癌癌因性疲乏裸鼠骨骼肌内Drp1表达,上调Mfn1表达,并且升高骨骼肌内ATP及GSH含量,降低MDA含量。结论:肺复康方能够有效缓解CRF症状,其机制可能与兴奋HPA轴,促进CORT分泌、抑制ACTH合成,促进骨骼肌融合,抑制其分裂,减少其氧化损伤有关。
Objective To investigate the effect of lung Fukang prescription on skeletal muscle of lung cancer-related fatigue nude mice.Methods 25 nude mice were randomly divided into blank control group,model group,low,medium and high dose group,Five in each group.Except for the blank control group,All four groups of nude mice were injected subcutaneously Lewis lung cancer cells,To establish a model of cancer-related fatigue in nude mice,Fatigue was measured by mouse tail suspension test.After three weeks of intervention,Five days a week,At the same time,according to the rat tail suspension experiment(2 times/week),the fatigue relief was judged.Six weeks after the drug intervention ended,The skeletal muscle of nude mice was taken for the dynamin-related protein(Drp1),mitofusins 1(Mfn1),ATP,malondialdehyde(MDA)and glutathione(GSH)detection.Results Before the intervention,compared with the blank control group,Both the model group and the dose group were prolonged.After drug intervention,lung Fukang prescription can down-regulate the expression of skeletal muscle Drp1 in lung cancer-related fatigue nude mice,By upregulating Mfn1 expression,To increase ATP and GSH levels in skeletal muscle,decrease MDA content.Conclusion Lung Fukang prescription can effectively relieve cancer-related fatigue symptoms,and its mechanism may be related to HPA axis of excitement,promote CORT secretion,inhibit ACTH synthesis,promote skeletal muscle fusion,inhibit its division,reduce its oxidative damage.
