牙周炎NLRP3炎性小体表达量与炎症反应、牙槽骨吸收的相关性研究

Study on the correlation between the expression of NLRP3 inflammatory factors and inflammatory response and alveolar bone resorption in periodontitis

目的:探究牙周炎NLRP3炎性小体表达量与炎症反应、牙槽骨吸收的相关性。方法:选取2015年5月~2018年5月我院收治的牙周炎患者127例为观察组,选取同期体检的127例健康人群为对照组。统计观察组细菌分布特征;比较2组NLRP3炎性小体mRNA的表达情况;比较2组龈沟液炎症因子水平及牙周炎的指标变化情况;对观察组NLRP3mRNA表达量与龈沟液炎症因子、牙槽骨吸收进行pearson相关性研究。结果:127例患者中菌株122株;观察组NLRP3、IL-1β、IL-18、Caspase-1的mRNA表达量均高于对照组;观察组龈沟液ICAM-1、CXCL12、HSP60量均高于对照组;观察组患者PLI、BI、PD、AL均高于对照组;NLRP3mRNA与ICAM-1、CXCL12、HSP60、PLI、BI、PD、AL均呈正相关(r=0.511,0.602,0.525,0.484,0.344,0.286,0.311)。结论:牙周炎患者的牙周膜细胞中NLRP3 mRNA表达量与龈沟液炎症因子、牙槽骨吸收成正相关,高表达的NLRP3炎性小体能够加剧炎症反应及牙槽骨吸收。

Objective To investigate the correlation between the expression of NLRP3 inflammatory factors and inflammatory response and alveolar bone resorption in periodontitis.Methods 127 patients with periodontitis admitted to our hospital from May 2017 to May 2018 were selected as the observation group and 127 healthy people were selected as the control group.Statistical observation group bacterial distribution characteristics;compare the expression of NLRP3 inflammatory corpuscle mRNA in two groups;compare the level of inflammatory factors in gingival crevicular fluid and periodontitis indicators in two groups;Pearson correlation between the expression of NLRP3 and inflammatory factors in gingival crevicular fluid and alveolar bone resorption was studied in observation group.Results 122 strains of bacteria were isolated from 127 patients;the mRNA expressions of NLRP3,IL-1,IL-18 and Caspase-1 in the observation group were higher than those in the control group;ICAM-1,CXCL12 and HSP60 in the gingival crevicular fluid of the observation group were higher than those in the control group;PLI,BI,PD and AL in the observation group were higher than those in the control group;the expression of NLRP3 in the observation group was pearso with gingival crevicular fluid inflammatory factors and periodontitis indicators.N correlation analysis showed that NLRP3 was positively correlated with ICAM-1,CXCL12,HSP60,PLI,BI,PD and AL(r=0.511,0.602,0.525,0.484,0.344,0.286,0.311).Conclusion In periodontal ligament cells of patients with periodontitis,the expression of NLRP3 was positively correlated with gingival crevicular fluid inflammation factors and alveolar resorption.Highly expressed NLRP3 inflammatory bodies could aggravate the inflammatory response and alveolar bone resorption in the course of periodontitis.
