应用转录组测序分析运动和感觉神经再生时基因表达差异的研究

Analysis of the difference in gene expression between motor and sensory nerve regeneration by transcriptome sequencing

摘要 目的通过转录组测序筛选周围神经再生时运动、感觉神经再生相关的差异基因方法取雌性SD大鼠6只,一侧制作股神经损伤修复模塑,另一侧作为正常对照I个月后取两侧股神经肌支和皮支进行转录组测序,筛选出差异表达的基因,然后对K进行差异基因功能和通路显著富集性分析。结果股神经转录组测序结果显示再生的肌支共2098个差异基因(其中上调1006个,下调1092个),再生的皮支共1567个差异基因(其中上调727个,下调840个),,肌支差异基因主要富集于细胞分化、多细胞生物发展、炎性反应、细胞黏附等过程=皮支差异基因主要富集于代谢过程、侧支发芽的正向调控、离子迁移、脂肪酸生物合成等过程。结论周围神经运动和感觉神经再生时存在显著差异的基因表达和不同的信号通路。 Objective To screen differential genes related to motor and sensory nerve regeneration by transcriptome sequencing.Methods Six female Sprague Dawley rats were used to make femoral nerve injury repair model on one side and normal control on the other.One month later,the bilateral femoral nerve motor and sensory branches were taken for transcriptome sequencing,and the differentially expressed genes were screened out followed by GO and KEGG enrichment analyses to characterize the transcriptome profile.Results A total of 2098 differentially expressed genes were detected in the regenerated motor branches.Among these genes,1006 were up-regulated and 1092 were down-regulated.A total of 1567 differentially expressed genes were detected in the regenerated sensory branches.Among these genes,727 were up-regulated and 840 were down-regulated.The motor differential genes were mainly enriched in the rell differentiation,multicellular biological development,inflammatory response,and cell adhesion.The sensory differential genes were mainly enriched in metabolic processes,positive regulation of collateral sprouting,ion transport,fatty acid metabolic process.Conclusion There are significant differences in gene expression and signal pathways between peripheral nerve motor and sensory nerve regeneration.
