环状RNA在膀胱癌中的研究现状与展望

Research status and prospect of circular RNAs in bladder cancer

环状RNA(circRNAs)是一种单链闭环RNA分子,因其在各种疾病的发生和发展中发挥关键的调控作用而受到越来越多的关注。系统综述circRNAs在膀胱癌中的研究现状,可为其进一步研究提供参考。随着高通量测序技术的快速发展,膀胱癌中发现越来越多差异性表达的circRNAs,并且通过调控下游微小RNA或RNA结合蛋白进而在膀胱癌的发生发展中发挥重要作用,包括调控细胞增殖、肿瘤生长抑制、细胞周期阻滞、细胞凋亡、肿瘤侵袭、迁移、转移、血管生成和顺铂化疗耐药等。circRNAs的表达与膀胱癌的许多临床病理特征显著相关,包括肿瘤大小、数量,分期分级,淋巴结转移,远处转移和复发。此外,circRNAs可用于预测膀胱癌患者的预后,如总生存期、无病生存期和无进展生存期。在此,我们简要综述了circRNAs在膀胱癌中的生物学功能和机制,以及这些circRNAs在膀胱癌诊断、预后和靶向治疗方面的潜在临床应用。

Circular RNAs(circRNAs)are single-stranded,closed-loop RNA molecules that have attracted increasing attention due to their key regulatory roles in the occurrence and progression of various diseases.The research status of circRNAs in bladder cancer was systematically reviewed to provide reference for further research.The literature on circRNAs in bladder cancer in recent years was reviewed and systematically reviewed.With the rapid development of high throughput sequencing technology,more and more differently expressed circRNAs in bladder cancer were found,and through the regulation of the downstream miRNA or RNA binding protein which played an important role in the occurrence and development of bladder cancer,including regulation of cell proliferation,tumour growth suppression,cell cycle arrest,apoptosis,invasion,migration,metastasis,angiogenesis,and cisplatin chemoresistance.The expression of circRNAs was significantly associated with many clinicopathological features of bladder cancer,including tumor size,number,TNM stage and grade,lymph node metastasis,local recurrence and distant metastasis.In addition,circRNAs can be used to predict the prognosis of patients with bladder cancer,such as overall survival,disease-free survival,and progression-free survival.Here,we briefly review the biological functions and mechanisms of circRNAs in bladder cancer,as well as the potential clinical applications of these circRNAs in the diagnosis,prognosis and targeted therapy of bladder cancer.
