星形细胞上调基因-1和沉默信息调节因子2在膀胱尿路上皮癌组织的表达及其临床意义 被引量:4

摘要 目的:探讨膀胱尿路上皮癌组织中星形细胞上调基因-1(AEG-1)和沉默信息调节因子2(SIRT2)的表达,并分析两者表达水平与膀胱尿路上皮癌临床病理特征的关系。方法:收集大庆油田总医院2014年3月至2020年10月病理学确诊的82例膀胱尿路上皮癌组织标本和癌旁组织,采用免疫组织化学方法检测组织中AEG-1和SIRT2表达水平,结合研究对象临床资料行χ^(2)检验统计学分析。结果:膀胱尿路上皮癌组织中AEG-1阳性表达率[62.20%(51/82)]明显高于癌旁组织[15.85%(13/82)],差异有统计学意义(χ^(2)=37.003,P<0.01)。膀胱尿路上皮癌组织中SIRT2阳性表达率[43.90%(36/82)]明显低于癌旁组织[81.71%(67/82)],两者差异有统计学意义(χ^(2)=25.084,P<0.01)。AEG-1表达水平与膀胱尿路上皮癌组织学分化程度、浸润深度、淋巴结转移和TNM分期明显相关(χ^(2)=5.394、5.586、5.590,P<0.05),差异有统计学意义。SIRT2表达水平与膀胱尿路上皮癌浸润深度、淋巴结转移和TNM分期明显相关(χ^(2)=7.173、4.457、4.850,P<0.05),差异有统计学意义。结论:在膀胱尿路上皮癌组织中AEG-1蛋白表达上调、SIRT2蛋白表达下调,AEG-1和SIRT2有助于评估膀胱尿路上皮癌患者不良预后。 Objective To investigate the expression of astrocyte elevated gene-1(AEG-1)and silent-mating information regulator 2(SIRT2)in urothelial carcinoma(UTC)tissues of the bladder and their expression correlation with clinicopathological characteristics of UTC.Methods The expression levels of AEG-1 and SIRT2 in 82 cases of UTC tissues of the bladder and adjacent urothelial tissues pathologically confirmed from March 2014 to October 2020 on Daqing Oilfield General Hospital were examined by envision immunohistochemistry.The clinical data of these subjects were analyzed by Chi-square test.Results The positive rate of AEG-1 expression in UTC tissue of the bladder[62.20%(51/82)]was significantly higher than that in non-cancerous adjacent urothelial tissues of the bladder[15.85%(13/82)](χ^(2)=37.003,P<0.01).The positive rate of SIRT2 expression in UTC tissues of the bladder[43.90%(36/82)]was significantly lower than that in non-cancerous adjacent urothelial tissues of the bladder[81.71%(67/82)](χ^(2)=25.084,P<0.01).The expression levels of AEG-1 were related to histology grade,tumor node metastasis(TNM)stage and lymph node metastasis of UTC tissues of the bladder(respectively,χ^(2)=5.394,5.586,5.590,P<0.05).The expression levels of SIRT2 were related to invasion depth,lymph node metastasis and TNM stage in UTC tissues of the bladder(respectively,χ^(2)=7.173,4.457,4.850,P<0.05).Conclusion AEG-1 protein expression was up-regulated and SIRT2 protein expression was down-regulated in UTC tissues,and AEG-1 and SIRT2 can be used to evaluate poor prognosis in patients with UTC of the bladder.
