响应面优化水提法提取玉米须多糖的工艺研究 被引量:3

摘要 以玉米须为原料,采用水提醇沉法对玉米须多糖进行提取,通过Sevage法除去其中所含的蛋白质,苯酚-硫酸法测定多糖含量,以多糖提取率为评价指标,考察液料比、提取温度、水提次数、提取时间对玉米须多糖的影响,并用响应面法优化玉米须多糖的提取工艺。确定水提法提取玉米须多糖的最佳提取条件是液料比15.9∶1,提取温度84.5℃,水提次数2次,提取时间62 min,在此条件下所得的多糖提取率为4.28%。 Using corn silk as the raw material,the corn silk polysaccharide was extracted by water extraction and alcohol precipitation,the protein contained in it was removed by the Sevage method,the polysaccharide content was determined by the phenol-sulfuric acid method,and the polysaccharide extraction rate was used as an evaluation index to examine the ratio of liquid to material.The effect of extraction temperature,water extraction times and extraction time on corn silk polysaccharides,and the response surface methodology was used to optimize the extraction process of corn silk polysaccharides.It was determined that the optimal extraction conditions for water extraction method to extract corn silk polysaccharides were liquid-to-solid ratio 15.9∶1,extraction temperature 84.5℃,water extraction times twice,extraction time 62 min,and the extraction rate of polysaccharides obtained under these conditions was 4.28%.
