摘要
目的检测大鼠心搏骤停(CA)/心肺复苏(CPR)后胞红蛋白(CyGB)和高迁移率族蛋白Bl(HMGB1)分别在大脑海马回CA1区的表达及外周血清中的含量,以研究其对复苏后的评价及意义c方法90只健康SD大鼠,随机分为假手术(SO)组,CPR组及干预治疗(IVT)组,每组分3、12、24、48及72 h 5个观察时间点。窒息法制作大鼠CA/CPR模型。ELISA法测定血清CyGB和HMGB1的含量,采用TUNEL染色法检测海马回CA1区神经元的凋亡率,SABC法检测CyGB和HMGB1在海马回CA1区中的表达即平均光密度值(AOD)。结果海马回CA1区CPR组中的AOD:CyGB的表达在CPR后3 h明显降低,且随时间延长而增强,24 h达高峰,但48 h又降低;HMGB1的表达在3 h开始升高,CPR组分别与SO组和IVT组比较,差异均有统计学意义(P<0.01)。外周血清CPR组中:CyGB的含量变化与在海马回CA1区中的表达变化一致,呈正相关(r=0.883,P<0.01);HMGB1的含量与在海马回CA1区中的表达亦呈正相关(r=0.926,P<0.01),CPR组分别与SO组和IVT组比较差异均有统计学意义(P<0.01)。海马回CA1区中神经元的凋亡率与HMGB1表达的AOD呈正相关(r=0.621,P<0.05)。结论CyGB与HMGB1在大脑的表达与在外周血清中的含量是一致的,其中HMGB1变化与神经元的凋亡是一致的;在大鼠CA/CPR后,CyGB有一定脑保护作用,HMGB1参与CA/CPR后损伤。因此,通过检测外周血清中CyGB和HMGB1含量对心肺脑复苏效果与预后可能有量化价值。
Objective To detect the expression of Cytoglobin(CyGB)and High mobility group box-1 protein(HMGB1)in CA1 region of hippocampus and the contents of serum after cardiac arrest(CA)/Cardiopulmonary Resuscitation(CPR)in rats,in order to study its evaluation and significance after resuscitation.Methods 90 healthy SD rats were random-Ly divided into Sham operation(SO)group,CPR group and Intervention treatment(IVT)group.At 3,12,24,48 and 72 hours(h),the model of CA/CPR was established by asphyxia in rats.The contents of Serum CyGB and HMGB1 were determined by Elisa,the apoptotic rate of neurons in CA1 region of hippocampus was detected by TUNEL staining,and the expression of CyGB and HMGB1 in CA1 region of hippocampus was detected by SABC method.Results The expression of AOD and CyGB in the hippocampal CA1 region decreased significantly at 3 hours after CPR,increased with time,reached a peak at 24 hours,but decreased at 48 hours,and the expression of HMGB1 increased at 3 hours after CPR,compared with SO group and IVT Group,respectively,P<0.01.In the peripheral serum CPR group,the expression of CyGB was positively correlated with the expression of CyGB in hippocampal CA 1 region,r=0.883,P<0.01,and the expression of HMGB1 in hippocampal CA1 region was positively correlated,r=0.926,P<0.01,CPR Group was also significantly different from SO group and IVT Group,P<0.01.There was a positive correlation between the apoptosis rate of neurons and the AOD expression of HMGB1 in CA1 region of hippocampus,r=0.621,P<0.05.Conclusion The expression of CyGB and H1V1GB1 in brain is consistent with that in peripheral serum,and the change of HMGB1 is consistent with the apoptosis of neurons.After CA/CPR,CyGB has some protective effect on brain and HMGB1 is involved in CA/CPR injury.Therefore,detection of the levels of CyGB and HMGB1 in peripheral serum may be of quan-titative value in the effect and prognosis of cardiopulmonary cerebral resuscitation.
