摘要
目的:检测屏障自整合因子1(BANF1)在人结肠癌组织和癌旁组织中的表达,并探讨其临床意义。方法:收集60例结肠癌组织及其配对癌旁组织,采用Real-time PCR技术检测BANF1 mRNA的表达,采用免疫组织化学技术检测BANF1蛋白的表达,并分析BANF1蛋白表达水平与患者临床病理特征的关系。结果:BANF1 mRNA在结肠癌和癌旁组织中均有表达,表达量分别为2.730±0.538、0.953±0.179,差异有统计学意义(t=17.614,P<0.001);结肠癌组织中BANF1蛋白表达的阳性率为71.67%(43/60),癌旁组织BANF1蛋白表达阳性率为21.67%(13/60),两者相比较差异具有统计学意义(χ^(2)=30.13,P<0.01)。BANF1蛋白表达水平与结肠癌临床分期(χ^(2)=9.712,P<0.05)有关,而与患者性别、年龄大小、肿瘤分化程度、淋巴结是否转移无关(P>0.05)。结论:BANF1在结肠癌组织中高表达,且与结肠癌的发生、发展有关。
Objective:To detect the expression of barrier self-integrating factor 1(BANF1)in human colon cancer and adjacent normal tissues,and to explore its clinical significance.Methods:60 colon cancer specimens and their paracancerous normal tissue samples were collected.The expression of BANF1 mRNA was detected by real-time PCR,and the expression of BANF1 protein was detected by immunohistochemistry,and its relationship with clinical and pathological characteristics was analyzed.Results:The expression levels of BANF1 mRNA were 2.730±0.538 and 0.953±0.179,respectively in 60 cases of fresh colon cancer and adjacent normal tissues,and the difference was statistically significant(t=17.614,P<0.001).The positive rate of BANF1 protein expression in colorectal adenocarcinoma tissues was 71.67%(43/60),and the positive rate of BANF1 protein expression in adjacent normal tissues was 21.67%(13/60),the difference was statistically significant(χ^(2)=30.13,P<0.01).The expression of BANF1 protein in patients with colorectal adenocarcinoma was related to its clinical stage(χ^(2)=9.712,P<0.05),but not related to gender,age,degree of differentiation and lymph node metastasis(P>0.05).Conclusion:BANF1 is highly expressed in colon cancer patients and is related to the occurrence and development of colon cancer.
