禽腺病毒(C种,4型)精制卵黄抗体研制及免疫效果评价 被引量:3

Preparation of avian adenovirus (type C,serotype 4) refined yolk antibody and evaluation of its immune effect

摘要 为了评价以禽腺病毒(C种,4型)Fiber-2蛋白为免疫原制备精制卵黄抗体的免疫效果,本研究使用重组大肠杆菌BL21(DE3)-Fiber-2株诱导表达禽腺病毒(C种,4型)Fiber-2蛋白,蛋白纯化后制备免疫原,免疫产蛋鸡收获高免蛋,经提纯获得的精制卵黄抗体AGP效价达1∶8;将精制卵黄抗体注射SPF鸡进行免疫效果评价。结果表明,精制卵黄抗体经肌肉或皮下注射SPF鸡,注射后7d内可100%保护鸡群抵抗禽腺病毒(C种,4型)强毒株的攻击,另外强毒株人工感染后24 h内肌肉或皮下注射抗体1.0 mL/只,可为感染鸡提供90%~100%的保护。本研究表明,以原核表达制备的禽腺病毒(C种,4型)Fiber-2蛋白为免疫原制备的精制卵黄抗体免疫效果较为理想,为后续抗体的临床应用提供了技术参考,并为该病的防控提供了新的思路。 In order to evaluate the preventive and therapeutic effects of the purified egg yolk antibody of flow adenovirus(type C,serotype 4) which was prepared with Fiber-2 protein as the immunogen, recombinant E.coli BL21(DE3)-Fiber-2 strain was used to induce the expression of Fiber-2 protein.High immunity eggs were obtained by immunizing laying hens with subunit vaccine prepared by purified protein, the purified egg yolk antibody AGP titer obtained reached 1∶8.The results of the prevention evaluation showed that the refined egg yolk antibody injected into SPF chickens by intramuscular or subcutaneous can 100% protect the chickens from the attack of the virulent avian adenovirus(type C,serotype 4) strain within 7 days after injection, and intramuscular or subcutaneous injection of antibody 1.0 mL/head 24 h after infection can provide 90% to 100% protection for chickens artificially infected with the virulent strains.This study shows that the immune effect of the purified egg yolk antibody prepared by using the avian adenovirus(type C,serotype 4) Fiber-2 protein is ideal, which provides technical reference for the clinical application of subsequent antibodies and provides new ideas for the prevention and control of the disease.
