鸭源沙门菌三价油乳剂灭活疫苗的制备及免疫效果

摘要 以江苏徐州地区主要流行的印第安纳沙门菌、阿贡纳沙门菌和鸭沙门菌3株鸭源沙门菌为研究对象,采用高密度发酵制备多价油乳剂灭活疫苗,检验疫苗性状并对疫苗免疫后雏鸭血清抗体水平的消长规律和淋巴细胞的活性进行检测,评价疫苗对雏鸭的免疫保护效果,以期制定出合理的免疫程序。结果显示,制备的疫苗为油包水剂型,物理性状稳定,无菌检测合格。雏鸭免疫抗体水平监测结果显示,单次免疫,抗体水平在免疫后3周达到高峰;多次免疫,抗体水平在免疫后5周达到高峰,7周后依然维持较高水平。E玫瑰花环试验检测T淋巴细胞数量,免疫组明显高于对照组。动物免疫试验结果显示,制备的三价油乳剂灭活疫苗可以有效抵抗强毒菌株的攻击,动物保护率达到87%以上。本研究制备的鸭源沙门菌多价油乳剂灭活疫苗性状稳定,免疫效果好,能有效抵抗强毒菌株攻击,为鸭沙门菌病的预防提供有效的防控生物制品。 In order to improve the ability of prevention and control of Salmonellosis in ducks, the experiment was conducted to development an effective vaccine.In this research, the multivalent oil-emulsion inactivated vaccine of Salmonella was prepared by high-density fermentation of the main Salmonella strains in Xuzhou.The vaccine characteristics were investigated, serum antibody level and lymphocyte activity were monitored, immunoprotective effect was conducted on the ducklings.The vaccine was stable water-in-oil emulsion.The results showed that the antibody level reached the peak at 3 weeks after single immunization, reached the peak at 5 weeks after multiple immunizations, and remained at a high level after 7 weeks.The E rosette forming rate in the groups were higher than that of the control group.The vaccine showed more than 87% protection rate, and could effectively resist the attack of virulent strains.All the results that the multivalent oil-emulsion inactivated vaccine are stable, long shelf life and good immune effects, and can very effectively resist the infection of virulent strains.The vaccine can provide effective prevention and control measures for the prevention of Salmonellosis in ducks in actual production, which has important practical significance.
