摘要
子宫内膜修复和前列腺素F_(2α)(PGF_(2α))分泌之间存在正相关关系,但PGF_(2α)在子宫内膜修复中发挥的具体作用目前仍未知。本试验通过体外培养奶牛子宫内膜组织,研究激活PGF_(2α)受体(prostaglandin F_(2α) receptor, PTGFR)对参与子宫内膜修复的一系列生长因子表达的影响。结果显示,在使用氟前列醇(fluprostenol)激活PTGFR后,子宫内膜组织中结缔组织生长因子(CTGF)、成纤维细胞生长因子-2(FGF-2)、转化生长因子-β1(TGF-β1)和血管内皮生长因子(VEGFA)的mRNA和蛋白表达水平均显著上调(P<0.05)。此外,子宫内膜组织中能够代表细胞发生增殖的增殖细胞核抗原(PCNA)水平也出现显著上升(P<0.05)。结果表明,PGF_(2α)-PTGFR通路的激活可通过诱导生长因子的表达对奶牛子宫内膜的生长起到促进作用。
A positive correlation between endometrial repair and prostaglandin F_(2α)(PGF_(2α)) secretion has been reported, but until now little was known about the roles that PGF_(2α) play in endometrial repair.In the present study, the regulatory roles of fluprostenol induced PGF_(2α) receptor(PTGFR) activation on the expression of a series of growth factors involved in endometrial repair were investigated in bovine endometrial explants in vitro.The results indicated that after the PGF2 receptor was activated by fluprostenol, the mRNA and protein expression of connective tissue growth factor(GTGF),fibroblast growth factor 2(FGF-2),transforming growth factor β1(TGF-β1) and vascular endothelial growth factor A(VEGFA) was increased in bovine endometrial explants(P<0.05).In addition, the level of proliferating cell nuclear antigen(PCNA) which can represent cell proliferation, was also significantly increased in endometrial explants(P<0.05).All these results suggest that the activation of PGF_(2α)-PTGFR pathway may promote endometrial repair in dairy cows through inducing growth factor expression.
