云南新平蔗区发现甘蔗白条病

Occurrence of Sugarcane Leaf Scald in Sugarcane Planting Area of Xinping, Yunnan Province, China

利用白条黄单胞菌[Xanthomonas albilineans(Ashby)Dowson]特异性引物XAF1/XAR1对2019年采自云南新平蔗区的8份疑似甘蔗白条病样品进行PCR分子检测,结果表明所有样品均扩增得到大小约608 bp的特异性条带,GenBank登录号为MT625864~MT625871。这些核苷酸序列与白条黄单胞菌(X.albilineans)GPE PC73菌株基因组上的三磷酸腺苷结合盒(Adenosine triphosphate-biding cas⁃sette,ABC)转运蛋白基因ab(c基因号XALc_1791)的序列一致性为100%。系统发育树分析表明这些白条黄单胞菌菌株序列处于同一进化分支。结合田间症状诊断和分子鉴定结果,确认云南新平蔗区发生的甘蔗病害为X.albilineans引起的甘蔗白条病,这是云南新平蔗区首次发现。

Using Xanthomonas albilineans(Ashby)Dowson specific primers of XAF1/XAR1 experiment PCR molecular detection for 8 suspected leaf scald samples collected from the Xinping of Yunnan Province in 2019.The results showed that all samples were amplified to a specific band of about 608 bp in size.GenBank accession number is MT625864-MT625871.These nucleotide sequences are 100%identical to the sequence of the adenos-ine triphosphate-biding cassette transporter gene(abc,gene number XALc_1791)in Xanthomonas albilineans GPE PC73 strain genome.Phylogenetic analysis showed that these Xanthomonas albilineans strains sequences were clustered together into one branch.Base on the result of symptom diagnosis in the field and molecular identi-fication,the sugarcane disease occurred in Xinping was confirmed to be leaf scald caused by Xanthomonas albi-lineans.This is the first occurrence of sugarcane leaf scald disease in Xinping area of Yunnan Province.
