液相芯片技术在花生品种纯度鉴定中的应用 被引量:1

摘要 本研究采用花生10K液相芯片结合Illumina测序平台进行高通量测序,检测100份花生样本,获得目标区域内总计1万个SNP。与表型鉴定结果完全一致。使用R、PLINK、MEGA-X等软件处理SNP数据,主成分分析、降维分析和聚类分析结果均表明,花生品种“东北王”的纯度为98.9%,本研究结果表明液相芯片技术在品种纯度鉴定上具有信息量大、灵敏度高和准确性极高等特点,为育种者鉴定花生品种纯度提供了新的选择。 Peanut 10K liquid chip and Illumina high-throughput sequencing technology were used for identification of Dongbeiwang cultivar purity in this study.A total of 100 peanut samples were detected and a total of 10000 SNP(Single Nucleotide Polymorphism)loci in the target regions were obtained.These SNP data were processed by R,PLINK,MEGA-X softwares.The results from principal components analysis,multidimensional scaling and cluster analysis showed that the purity of peanut Dongbeiwang detected was 98.9%,which was completely consistent with that using phenotype identification.The results indicated that liquid chip technology has characteristics of abundant genetic information,high sensitivity and high accuracy for identification of varietal purity,which provided a new choice for breeders to identify varietal purity of peanut.
