摘要
目的观察miRNA-132在不同骨肉瘤细胞系中的表达及对骨肉瘤细胞生物学行为的影响,探讨miRNA-132在人骨肉瘤发生发展中的作用。方法通过荧光定量PCR技术检测不同骨肉瘤细胞系U2OS、MG63、143B中miRNA-132的表达情况,使用pRFP-miRNA-132-down质粒转染U2OS细胞、pRFP-miRNA-132-up质粒转染143B细胞,使用EdU检测肿瘤细胞增殖状态,Transwell小室检测肿瘤细胞迁移和侵袭能力。结果在3种骨肉瘤细胞系U2OS、MG63、143B中,miRNA-132的表达依次下降(U2OS>MG63>143B);降低miRNA-132在U2OS细胞中的表达促进细胞的增殖和侵袭能力,增加miRNA-132在143B细胞中的表达抑制细胞的增殖和侵袭能力。结论miRNA-132可能通过抑制肿瘤细胞的增殖和侵袭调节骨肉瘤的发生发展。
Objective To observe the expression of miRNA-132 in different osteosarcoma cell lines and its influence on biological behaviors of osteosarcoma cells, and to discuss its effect on the occurrence and development of osteosarcoma. Methods Detect the expression of miRNA-132 in U2OS, MG63 and 143B cell lines by RT-qPCR. Transfect U2OS cells with pRFP-miRNA-132-down plasmid and 143B cells with pRFP-miRNA-132-up plasmid. Detect the proliferation of tumor cells via EdU staining. Detect the migration and invasion capacity of tumor cells via transwell chamber and detect the cellular apoptosis state with TUNEL kit. Results The U2OS, MG63 and 143B cell lines showed varing levels of miRNA-132 expression(U2OS>MG63>143B);decreasing the expression of miRNA-132 in U2OS cells enhanced cellular migration and invasion capacity, while increasing its expression in 143B cells inhibited cellular migration and invasion capacity. Conclusion mi RNA-132 might regulate the occurrence and development of osteosarcoma by inhibiting the proliferation and invasion of tumor cells.
