摘要
目的探索着丝粒蛋白U(centromere protein U,CENPU)在卵巢癌组织中表达情况和与卵巢癌细胞增殖和迁移的关系及其相关机制。方法应用免疫组织化学和Western blot检测卵巢癌组织和癌旁组织、卵巢癌细胞和正常卵巢上皮细胞中CENPU的表达;沉默卵巢癌细胞CENPU后,应用CCK-8、Transwell和划痕实验检测卵巢癌细胞增殖、侵袭和迁移能力的变化,以Western blot检测p-PI3K和p-AKT的表达情况。结果相对于癌旁组织,卵巢癌组织中CENPU显著上调,且其表达与FIGO分期、肿瘤大小和淋巴结转移呈正相关。相对于正常卵巢上皮细胞,卵巢癌细胞中CENPU显著上调。敲低CENPU显著抑制卵巢癌细胞增殖、迁移、侵袭,以及p-PI3K和p-AKT的表达。结论CENPU在卵巢癌中表达上调;CENPU可能通过PI3K/AKT信号通路促进卵巢癌细胞的增殖和转移。
Objective To explore the expression of centromere protein U(CENPU)in ovarian cancer tissues,and its relationship with proliferation and migration of ovarian cancer cells as well as its related mechanisms.Methods The expression level of CENPU in ovarian cancer tissues and paracancer normal tissues was detected by immunohistochemistry staining,and its expression in ovarian cancer cells and normal ovarian epithelial cells was detected by Western blot.After CENPU silencing,the proliferation,invasion and migration of ovarian cancer cells were detected by CCK-8,Transwell and scratch wound assays,respectively.And the expression levels of p-PI3K and p-Akt were detected by Western blot.Results CENPU was significantly up-regulated in ovarian cancer tissues compared with paracancerous tissues,and its expression was positively correlated with FIGO stage,tumor size and lymph node metastasis.CENPU was significantly up-regulated in ovarian cancer cells compared with normal ovarian epithelial cells.Knockdown of CENPU significantly inhibited proliferation,migration and invasion of ovarian cancer cells,as well as the expression levels of p-PI3K and p-Akt.Conclusions CENPU expression was up-regulated in ovarian cancer.CENPU may promote the proliferation and metastasis of ovarian cancer cells by PI3K/Akt signaling pathway.
