摘要
目的检测神经纤毛蛋白1(neuropilin 1,NRP1)在卵巢癌组织中的表达水平与病理特征的关系,为其治疗和预后提供理论指导。方法选取2013年至2019年于西安交通大学第一附属医院治疗的120例卵巢癌患者及配对的癌旁组织23例,利用实时荧光定量PCR、蛋白免疫印迹分析、免疫组织化学染色分别检测NRP1在卵巢癌组织中的mRNA及蛋白表达水平,并分析NRP1免疫反应性与临床病理特征的关系。同时利用免疫荧光检测NRP1在卵巢癌细胞系SKOV3中的表达水平。结果NRP1蛋白在卵巢癌组织中表达水平显著高于癌旁组织,在卵巢癌组织中阳性表达率(76.67%)明显高于癌旁组织(47.83%)。NRP1的高阳性表达率与肿瘤组织低分化相关,与肿瘤组织高分期相关,与淋巴结转移及CA125血清表达水平均呈正相关。NRP1在卵巢癌细胞系中表达水平也较高。结论NRP1蛋白在卵巢癌中高表达,与卵巢癌的病理分级分期密切相关,可作为一个预后指标指导卵巢癌治疗。
Objective To investigate the expression of neuropilin 1(NRP1)in human ovarian cancer tissues and its correlation with the clinicopathologic parameters,to provide guidance for the treatment and prognosis of ovarian cancer.Methods The cancer tissues of 120 patients with ovarian cancer and treated in The First Affiliated Hospital of Xi’an Jiaotong University from 2013 to 2019,as well as 23 cases of paired adjacent non-tumor tissues were selected.Real-time fluorescent quantitative PCR was used to detect the mRNA expression level of NRP1.Western blotting and immunohistochemistry assay were performed to detect the protein expression of NRP1 in ovarian cancer tissues.Then we analyzed the relationship between NRP1 immunoreactivity and clinicopathologic parameters.Meanwhile,immunofluorescence assay was performed to examine the expression of NRP1 in the ovarian cancer cell line SKOV3.Results The NRP1-immunoreactivity in cancer tissues was significantly higher than that in paired adjacent non-tumor tissues,and the positive rate of NRP1 in ovarian cancer(76.67%)was also significantly higher than that in paired adjacent non-tumor tissues(47.83%).The high rate of NRP1 was correlated with the poor differentiation of tumor cells and high tumor FIGO stage,and was positively correlated with lymph node metastasis and serum level of CA125.NRP1 is also highly expressed in SKOV3 ovarian cancer cells.Conclusion NRP1 is highly expressed in ovarian cancer tissues.Its expression is closed related to the pathological grading and staging of tumor,and it can be used as a prognostic index in the treatment of ovarian cancer.
