两种操作方法在EBER原位杂交染色中的比较与应用

Comparison and application of two different operation methods in EBER in situ hybridization staining

目的比较全自动免疫组织化学染色机染色和手工染色在EBER原位杂交技术中的应用。方法收集EBER染色阳性表达的不同组织标本35例,每例切片2张,分别采用罗氏Ventana免疫组织化学染色机染色和手工染色两种方式进行EBER原位杂交染色,对染色结果进行比较和分析。结果35例手工组与仪器组染色结果一致,但仪器组阳性细胞着色深,与背景染色对比鲜明,且仪器组节省时间11h。结论两种方法在EBER染色中结果一致,但仪器组图像质量更好,且操作方法简便,更适合临床推广。

Objective To compare the two methods of automated immunohistochemistry staining machine aided staining and manual staining during EBER CISH operation.Methods 35 samples of different tissues with EBER positive expression were collected,2 sections for each case.EBER in situ hybridization staining was performed by two methods,i.e.Roche Ventana immunohistochemical staining machine aided staining and manual staining,and the results were compared and analyzed.Results The results of 35 cases were consistent in both two group,but the positive stained cells in the instrument aided group have deep staining,sharping contrast with the background,and the instrument aided group save about 11 operating hours.Conclusion The staining effects of the two operation methods are consistent in EBER staining,but the instrument aided methods provids images with higher quality and is simple to operate,therefore is worthy of clinical promotion.
