摘要
目的利用生物信息学分析探讨线粒体基因在前列腺癌生化复发中的作用。方法利用TCGA及BROAD数据分析前列腺癌与生化复发相关的线粒体基因;网站VENNY、STRING、METASCAPE及软件Cytoscape分析相关基因在前列腺癌生化复发中发挥作用的相关信号通路。结果筛选出141个与前列腺癌生化复发相关的线粒体基因。其中相互作用最密切的前20个基因分别是SOD2、CS、COX4I1、ECHS1、HMGCL、PTCD3、CPT2、NDUFA9、HIBADH、ALDH6A1、PARK7、PCCA、NDUFV2、MRPL17、DAP3、ACSF3、AURKAIP1、MRPL20、MDUFA13及MRPL40。这些线粒体基因主要参与线粒体基质和线粒体膜的组成,参与有机酸、脂肪酸的代谢,调控氧化还原活动。结论线粒体基因对前列腺癌生化复发具有影响作用,可能是前列腺癌的早期诊断及靶向治疗的潜在方向。
Objective To investigate the role of mitochondrial genes in biochemical recurrence of prostate cancer by bioinformatics analysis.Methods Dataset from TCGA and BROAD were downloaded to analyze the genes related to biochemical recurrence of prostate cancer and mitochondrial genes.The roles of genes related to biochemical recurrence of prostate cancer were analyzed by VENNY,STRING,METASCAPE website and Cytoscape software.Results There were 141 mitochondrial genes associated with biochemical recurrence of prostate cancer.The top 20 genes were SOD2,CS,COX4I1,ECHS1,HMGCL,PTCD3,CPT2,NDUFA9,HIBADH,ALDH6A1,PARK7,PCCA,NDUFV2,MRPL17,DAP3,ACSF3,AURKAIP1,MRPL20,MDUFA13 and MRPL40.These mitochondrial genes are mainly involved in the composition of mitochondrial matrix and mitochondrial membrane,the regulation of organic acid and fatty acid metabolism,and the modulation of oxidoreductase activities.Conclusion The mitochondrial genes play an important role in the biochemical recurrence of prostate cancer,which may be a potential direction for early diagnosis and targeted treatment of prostate cancer.
