摘要
目的:观测小鼠磨牙加力不同时间点脾脏CD3^(+)、CD8^(+)、CD4^(+)T淋巴细胞及Th细胞亚群的比例变化,分析正畸加力对免疫系统中T淋巴细胞变化的影响。方法:4周龄雄性C57BL/6小鼠随机分为1 d、3 d、5 d和7 d加力组以及相应时间的对照组。加力组向磨牙施加25 g力;对照组安置相同加力装置,力值为0。使用流式细胞术检测小鼠脾脏单个核细胞中CD3^(+)比例、CD4^(+)/CD3^(+)比例、CD8^(+)/CD3^(+)比例及Th1,Th2,Th17细胞占CD4^(+)细胞比例,以及CD3,CD4,CD8,IFN-γ,IL-4,IL-17荧光强度的变化。结果:与对照组相比,7 d实验组CD8^(+)T细胞占CD3^(+)T细胞的比例和Th1细胞占CD4^(+)T淋巴细胞的比例显著增高(P<0.05);CD3,CD8,IFN-γ,IL-17平均荧光强度均显著增高(P<0.05);相反,CD4^(+)T细胞占CD3^(+)T淋巴细胞的比例显著降低(P<0.01)。而1,3,5 d实验组与对照组间未见明显差异(P>0.05)。结论:正畸加力过程可引起小鼠脾脏T细胞亚群占比和及其分泌的细胞因子发生变化。
Objective:To analyze the effects of orthodontic treatments on the changes of T lymphocytes in immune systems,by observing the changes of the proportions of CD3^(+),CD8^(+),CD4^(+)T lymphocytes and Th cell subsets in the spleens of mice at different time points during orthodontic treatments in mice.Methods:Male C57BL/6 mice aged 4 weeks were randomly divided into 1,3,5,7 days orthodontics loading groups and corresponding control groups.Mice in orthodontics loading groups were delivered 25 g of force.Mice in control groups were placed the same devices as orthodontics loading groups,but the force value was 0 g.Flow cytometry was used to detect the changes of fluorescence intensities of different cytokines containing CD3,CD4,CD8,IFN-γ,IL-4,IL-17 and several ratios in spleen mononuclear cells of mice as following,the ratio of CD3^(+)cells to T lymphocytes,the respective ratios of CD4^(+)and CD8^(+)cells to CD3^(+)cells,as well as the respective ratios of Th1,Th2 and Th17 cells to CD4^(+)cells.Results:Compared with the control group,the proportion of CD8^(+)T cells in CD3^(+)T cells and the proportion of Th1 cells in CD4^(+)T lymphocytes in the 7-day-experimental group significantly increased(P<0.05).At the same time,the mean fluorescence intensities of CD3,CD8,IFN-γand IL-17 significantly increased(P<0.05).In contrast,the proportion of CD4^(+)T cells to CD3^(+)T lymphocytes significantly reduced(P<0.01).On day 1,3 and 5,there was no significant differences between the experimental groups and the control groups(P>0.05).Conclusion:The proportions of T cell subsets and the expression levels of cytokines secreted in the spleens of mice changed during orthodontic treatments.
