摘要
目的建立双柱双检测器顶空进样气相色谱法测定血中乙醇含量的方法。方法采用全自动进样器经一个进样口进样,分流到2个性质不同的色谱柱,再通过2个独立的氢火焰离子化检测器(FID)对样品进行分析。结果血中乙醇含量浓度在5 mg/100 ml~300 mg/100 ml时线性关系较好,相关系数均达到0.9999。方法的检出限和定量限分别为0.05 mg/100 ml、0.2 mg/100 ml和0.09 mg/100 ml、0.3 mg/100 ml,相对标准偏差分别为1.04%~3.02%和1.13%~3.47%,回收率分别为89.3%~97.8%和96.8%~98.4%。结论本方法满足国家标准方法要求,简便、快捷,提高检测效率,避免气相色谱只凭保留时间对复杂化合物中未知组分做出不可靠定性的缺点,适用于基层实验室批量检测血中乙醇。
Objective To establish a method for determination of ethanol content in blood by headspace injection gas chromatography with dual column and dual detector.Methods An automatic sampler was used to inject the samples through an injector,and the samples were separated into two different columns.The samples were then analyzed by two independent hydrogen flame ionization detectors.Results The blood ethanol concentration in the range of 5 mg/100 ml-300 mg/100 ml had a good linear relationship,and the correlation coefficients all reached 0.9999.The detection limits and quantitation limits were 0.05 mg/100 ml,0.2 mg/100 ml,0.09 mg/100 ml,and 0.3 mg/100 ml,the relative standard deviations were within 1.04%-3.02% and 1.13%-3.47% respectively.The recovery rates were within 89.3%-97.8% and 96.8%-98.4%.Conclusion This method meets the requirements of the national standard method,which is simple,fast,and improves the detection efficiency.It avoids the shortcoming that gas chromatography only makes unreliable qualitative determination of unknown components in complex compounds based on the retention time,and is suitable for the batch detection of ethanol in blood in the grass-roots laboratory.
