Quantitative Determination of Xanthoceras sorbifolium in Different Harvest Periods by HPLC

摘要 目的基于多元统计学分析及多成分含量测定,对不同生长期的文冠木药材进行质量评价,探讨文冠木适宜采收期。方法建立HPLC测定文冠木中4种黄酮类成分含量的方法,结合指纹图谱、聚类分析(HCA)和主成分分析(PCA)综合评价不同生长期文冠木药材的质量,并进一步经偏最小二乘法-判别(PLS-DA)筛选差异成分。结果不同生长期文冠木总化学成分呈动态变化,在S11~S20期间整体呈上升趋势,S41~S55期间增长为最迅速,总化学成分含量在S51~S55时达到最大值;指纹图谱样品相似度均大于0.90;HCA和PCA分析将样品分成4类;经过PLS-DA分析筛选出了二氢槲皮素和表儿茶素等7个差异成分。结论本实验所建立的文冠木HPLC指纹图谱和多成分含量测定的方法准确可行,根据文冠木不同生长期的化学成分动态累积变化及综合评价结果确定11月可能为最佳适宜采收期。 OBJECTIVE To evaluate the quality of Xanthoceras sorbifolium in different growth stages based on multivariate statistical analysis,and discuss the suitable harvesting period of Xanthoceras sorbifolium.METHODS An HPLC method was established for the determination of four flavonoids(epicatechin,dihydromyricetin,dihydroquercetin and myricetin)in the medicinal materials of the Xanthoceras sorbifolium.Fingerprinting,cluster analysis(HCA)and principal component analysis(PCA)were used to comprehensively evaluate the quality of the medicinal materials of different growth stages,and the differential components were further screened by partial least squares-discrimination(PLS-DA).RESULTS The total chemical composition of Xanthoceras sorbifolium in different growth stages showed a dynamic change.During the period from S11 to S20,the whole process showed an upward trend.During the S41-S55 period,the growth rate was the fastest,and the total chemical composition reached the maximum in November.The similarity of fingerprint samples was greater than 0.90.HCA and PCA analysis divided the medicinal materials of different growth stages into three categories.Seven different components such as ciprofloxacin and epicatechin were screened by PLS-DA analysis.CONCLUSION The fingerprint method and multi-component determination method of Xanthoceras sorbifolium are accurate and feasible.According to the dynamic accumulation change of chemical composition in Xanthoceras sorbifolium of different growth stages and the comprehensive evaluation results,the suitable harvesting period is November.
