摘要
目的观察推拿按法对腰椎间盘突出症大鼠步态行为及背根神经节白细胞介素(IL)-23及其受体(IL-23R)蛋白表达的影响,探讨其治疗腰椎间盘突出症的作用机制。方法将24只雄性SD大鼠随机分成正常组、模型组、推拿组,每组8只。采用坐骨神经慢性压迫性损伤模型模拟腰椎间盘突出症。推拿组于成模后第4~17日予推拿按法干预,每日1次,每次10 min。测量大鼠右后足步态数据,HE染色观察背根神经节形态,免疫荧光染色和Western blot检测背根神经节IL-23、IL-23R蛋白的表达。结果与正常组比较,模型组大鼠右后足最大接触面积、足印面积显著减少,摆动时间明显增加(P<0.01);背根神经节出现不同程度坏死、炎性细胞浸润,IL-23、IL-23R蛋白表达明显升高(P<0.05)。与模型组比较,推拿组大鼠第17日右后足最大接触面积、足印面积增加,摆动时间减少(P<0.05);背根神经节出现坏死、炎性细胞浸润,IL-23、IL-23R蛋白表达明显降低(P<0.05)。结论推拿按法能改善腰椎间盘突出症大鼠步态行为,其机制可能与抑制背根神经节IL-23、IL-23R蛋白的表达,改善外周敏化有关。
Objective To observe the effects of massage pressure on gait behavior and expressions of IL-23 and IL-23R of dorsal root ganglia in rats with lumbar disc herniation;To explore the mechanism of its treatment of lumbar disc herniation.Methods Totally 24 male SD rats were randomly divided into normal groups,model group,and massage group,with 8 rats in each group.The chronic constriction injury of the sciatic nerve in rats was used to establish the neuropathic pain model.Massage group received the massage pressure intervention once a day for 10 minutes from the 4th to the 17th day after modeling.The gait data of the right hindfoot of rats were measured,and the morphological changes of the dorsal root ganglia were observed by HE staining;the expressions of IL-23 and IL-23R protein in the dorsal root ganglia were detected by immunofluorescence staining and Western blot.Results Compared with the normal group,the max contact area and the print area of the right hindfoot of the model group significantly decreased,and the swing time significantly increased(P<0.01);the dorsal root ganglia appeared necrosis,inflammatory cell infiltration,and the expressions of IL-23 and IL-23R increased significantly(P<0.05).Compared with the model group,the max contact area and the print area of the right hindfoot of the massage group on the 17th day increased,and the swing time decreased(P<0.05);the dorsal root ganglia appeared necrosis,inflammatory cell infiltration,and the expressions of IL-23 and IL-23R decreased significantly(P<0.05).Conclusion The massage pressure can improve the gait behavior of rats with lumbar disc herniation,and its mechanism may be related to inhibiting the expressions of IL-23 and IL-23R in dorsal root ganglia and improving peripheral sensitization.
