长链非编码RNA PGM5-AS1在结直肠癌中的临床意义及功能研究

摘要 目的探讨长链非编码RNA PGM5-AS1在结直肠癌组织中的表达情况及其对结直肠癌细胞增殖、迁移的影响。方法通过实时荧光定量PCR检测24例结直肠癌组及癌旁组、正常人结直肠上皮细胞系NCM460及结直肠癌细胞系SW480、HCT116、Lovo中PGM5-AS1 mRNA的表达。体外构建PGM5-AS1过表达载体,转染结直肠癌细胞SW480、HCT116,构建PGM5-AS1过表达细胞模型,采用荧光定量PCR、CCK-8、细胞划痕实验检测PGM5-AS1水平、增殖活力、划痕愈合率。结果相比癌旁组织,PGM5-AS1在结直肠癌组织中表达降低(t=11.41,P<0.05)。3种结直肠癌细胞系(SW480、HCT116、Lovo)PGM5-AS1的表达较NCM460细胞低(t分别=6.76、7.28、9.47,P均<0.05)。HCT116、SW480细胞中,过表达组的PGM5-AS1 mRNA相对表达量高于空载组(t分别=5.72、3.47,P均<0.05)。过表达组转染48 h、72 h、96 h后的细胞增殖活力低于对照组和空载组(t分别=11.72、13.64;7.47、9.67;9.28、7.41,P均<0.05);过表达组的划痕愈合率低于对照组和空载组,差异有统计学意义(t分别=7.73、5.26,P均<0.05)。结论结直肠癌细胞中PGM5-AS1表达降低,上调PGM5-AS1水平可抑制细胞增殖、迁移能力,可能通过抑制细胞增殖来发挥抑癌功能,成为结直肠癌治疗的潜在靶点。 Objective To investigate the expression of long noncoding RNA PGM5AS1 in colorectal cancer tissues and its effect on the proliferation and migration of colorectal cancer cells.Methods The expression of PGM5AS1 mRNA was detected by realtime quantitative PCR in 24 cases of colorectal cancer and adjacent,normal colorectal epithelial cell lines NCM460,colorectal cancer cell lines SW480,HCT116,and Lovo.PGM5AS1 overexpression vector was constructed in vitro and transfected into colorectal cancer cells SW480 and HCT116 to construct PGM5AS1 overexpression cell model.The level of PGM5AS1,proliferation activity and scratch healing rate were detected by qPCR,CCK8 and scratch assay.Results Compared with the tissue adjacent to carcinoma,the expression of PGM5AS1 decreased in colorectal cancer tissues(t=11.41,P<0.05).The expression of PGM5AS1 in three colorectal cancer cell lines(SW480,HCT116,Lovo)was lower than that in NCM460 cell lines(t=6.76,7.28,9.47,P<0.05).In HCT116 and SW480 cells,the expression level of PGM5AS1 in overexpression group was higher than that in noload group(t=5.72,3.47,P<0.05).After transfection for 48h,72h and 96h,the proliferation activity of overexpression group was lower than that of control group and noload group(t=11.72,13.64,7.47,9.67,9.28,7.41,P<0.05).The scar healing rate of the overexpression group was lower than that of the control group and the noload group(t=7.73,5.26,P<0.05).Conclusion The expression of PGM5AS1 in colorectal cancer cells is decreased.Upregulation of PGM5AS1 level could inhibit the ability of proliferation and migration of colorectal cancer cells,which may play a role in tumor suppression by inhibiting cell proliferation,and may be a potential target for the treatment of colorectal cancer.
