摘要
Foxl2 (winged helix/forkhead transcription factor gene 2)是叉头框转录因子家族成员之一,在鱼类性别决定与分化中起重要的作用。为了探讨foxl2基因在泰国斗鱼(Bettas plendens)雌雄组织中的表达差异,本研究利用分子克隆技术分离并鉴定了泰国斗鱼foxl2基因cDNA序列1 246 bp,其中开放阅读框为921 bp,共编码306个氨基酸。多重序列比对分析,泰国斗鱼Foxl2氨基酸序列与其他鱼类的Foxl2具有较高的同源性,显示出序列进化的保守性。组织表达结果显示,雌性泰国斗鱼foxl2基因在性腺中表达量最高;在雄鱼中,foxl2基因在性腺中表达量最低,结果表明foxl2基因组织表达具有显著的特异性及性别差异。本研究结果为后续开展泰国斗鱼的性别调控机理提供了参考。
Winged helix/Forkhead transcription factor gene 2(Foxl2) is a member of the forkhead box transcription factor family, plays an important role in sex determination and differentiation in fish. In order to explore the expression difference of foxl2 gene in the male and female tissues in Bettas plendens, the foxl2 cDNA was cloned and analyzed. In this study, the full-length ORF of foxl2 in B. plendens is 921 bp, encoding 306 amino acids.Multiple sequence alignment analysis revealed that the Foxl2 in B. plendens had high homology with other fish, suggesting that Foxl2 is conservative in teleost. The results of tissue distribution showed that the foxl2 gene is mainly expressed in gonad of females, and least expressed in gonad of males. The results showed that foxl2 tissue expression had significant specificity and sexual dimorphism. The research results provided an original basis for foxl2 in the sex determination and differentiation of B. plendens.
