1株表现为鞭毛抗原3相的韦太夫雷登沙门氏菌的鉴定和基因组序列分析

Genomic Sequence Analysis of a Salmonella enterica subsp. enterica serovar Weltevreden Strain with Triphasic Serovar

摘要 利用高通量测序获取1株抗原式为3,10:a, r, z6的沙门氏菌(Salmonella) GX150603的全基因组序列。根据鞭毛抗原序列、致病性和抗性基因预测GX150603的血清型,利用分子生物学软件分析基因组岛和前噬菌体,并与其他菌株进行全基因组系统发育分析。经鉴定GX150603为韦太夫雷登沙门氏菌(Salmonella enteric asubsp. enterica serovar Weltevreden),并推测由于鞭毛抗原的氨基酸变异导致该菌株与H:a血清产生了交叉反应。GX150603基因组携带148个毒力相关基因,8个耐药相关基因,27个基因组岛和3个前噬菌体。基因组岛包括沙门氏菌毒力岛SPI-1/2/3/4/5/6/9/11/13/14/19,centisome 63,CS54和3个该血清型特有的基因组岛。与7个韦太夫雷登沙门氏菌欧洲分离株比较,GX150603有4个共线性片段重排,4个共线性片段缺失和1个共线性片段存在较大差异,进化分析显示,同一血清型仍然表现出最近的亲缘关系。 Draft genome sequence of Salmonella GX150603 with antigen type 3,10: a, r, z6 was obtained based on high throughput sequencing technology. Serotype of GX150603 was predicted by flagellar genes sequence,pathogenicity and resistance genes, genomic islands and prophages were analyzed by molecular biology software,compared with other strains and genome-wide phylogenetic analysis was also carried out. GX150603 was identified as Salmonella enterica subsp. enterica serovar Weltevreden and serological reacted with H:a serum due to flagellar antigen variation. GX150603 carried 148 virulence factor genes, 8 antibiotic resistance genes, 27 genomic islands and 3 prophage Salmonella genomic islands include Salmonella pathogenicity island SPI-1/2/3/4/5/6/9/11/13/14/19, centisome 63 pathogenicity island,CS54 island and 3 serovar-specific genomic islands. Compared with to another 7 S. Weltevreden isolated in Europe, there were 4 presumably homologous blocks rearrangement,4 blocks deficiency and 1 block showing diversity in GX150603. Evolutionary analysis showed that the same serotype still showed the closest relatedness.
