通冠胶囊对ISO诱导大鼠心肌缺血的保护作用

Experimental study on the protective effect of Tongguan capsule on isoprenaline-induced myocardial ischemia in rats.

目的分析通冠胶囊改善心肌缺血的保护作用、量效关系及作用机制。方法此次实验采用皮下注射异丙肾上腺素(ISO,85 mg/kg)复制心肌缺血模型。实验分为现象实验、量效实验、机制实验3个方面。现象实验中,大鼠随机分为对照组(NE组)、低剂量模型组(ISO-L组)、高剂量模型组(ISO-H组)、通冠组(TGC组)、美托洛尔组(METO组)。量效实验中,大鼠随机分为NE组、ISO组、5个TGC组(剂量分别为0.01、0.05、0.1、0.5、1 g/kg)。机制实验中,大鼠随机分为NE组、ISO组、PD98059组(PD组)、LY294002组(LY组)、KN93组(KN组)、L-NAME组、TGC组;通过测定样本血液中心酶五项和肌钙蛋白结合解剖学分析TGC改善心肌缺血作用。结果现象实验结果显示模型组较对照组数据升高,METO组和TGC组较ISO组都有降低,但TGC组数据降低更为明显。量效实验结果显示ISO组较NE组数据高,随着给药剂量加大,数据先降低后有回升。机制实验结果显示ISO组较NE组数据升高,LY组、PD组、KN组较ISO组数据均有下降,L-NAME组与ISO组基本持平。结论通冠胶囊能改善大鼠心肌缺血现象且效果优于美托洛尔,最有效剂量是0.5 g/kg。通冠胶囊能显著减少心肌缺血损伤鼠是通过抑制eNOS通路,而不是通过AKT通路。

Objective To analyze the protective efficacy of Tongguan capsule(TGC)on myocardial ischemia,and its concentration-response relationship and mechanism.Methods The myocardial ischemia model was established by subcutaneous injection of isopropyl adrenaline(ISO,85 mg/kg)in rats.Experiment is divided into three aspects:phenomenon experiment,dose-effect experiment and mechanism experiment.In phenomenon experiment,the rats were randomly divided into control group(NE group),low dose(10 mg/kg)group(ISO-L group),high dose(85 mg/kg)group(ISO-H group),TGC(1 g/kg)group and metoprolol group(METO group)(10 mg/kg).In dose-effect experiment,the rats were randomly divided into NE group,ISO group,and five TGC groups(doses were 0.01,0.05,0.1,0.5,1 g/kg).In mechanism experiments,rats were randomly divided into NE group,ISO group,PD98059 group(PD group),LY294002 group(LY group),KN93 group(KN group),L-NAME group and TGC group.The calcium of muscle and heart enzyme in blood sample were to analyzed.Results Tongguan capsule can significantly improve myocardial ischemia of rats plasma,and the best dose was 0.5 g/kg.Tongguan capsule eNOS pathways through improving myocardial ischemia in rats.Conclusion Our result demonstrated that Tongguan capsule can improve myocardial ischemia in rats and the effect is better than metoprolol,the most effective dose is 0.5 g/kg.Tongguan capsule can significantly reduce the myocardial ischemia injury rats by inhibition of eNOS pathway,not the AKT pathway.
