摘要
目的建立不同产地茺蔚子饮片的超高效液相色谱(UPLC)指纹图谱,并评价饮片质量。方法色谱柱为Waters HSS T3柱(150 mm×2.1 mm,1.8μm),流动相为乙腈-0.1%三氟乙酸溶液(梯度洗脱),流速为0.30 m L/min,检测波长为254 nm,柱温为30℃。以4-羟基苯甲酸为参照,测定18批茺蔚子饮片的UPLC图谱,采用"中药色谱指纹图谱相似度评价系统(2012.0版)"软件进行相似度评价,确定共有峰,并对其进行聚类分析和主成分分析。结果18批茺蔚子饮片的UPLC图中有13个共有峰,相似度为0.801~0.982;18批茺蔚子饮片可聚为3类;峰4、峰10和峰12是判断不同产地茺蔚子饮片质量的重要因素。结论不同产地茺蔚子饮片的质量有显著差异。所建立的UPLC指纹图谱可为茺蔚子饮片的真伪鉴别和质量评价提供参考。
Objective To establish an ultra-performance liquid chromatographic(UPLC)fingerprints of Leonuri Fructus from different habitats,and evaluate its quality.Methods The chromatographic column was waters HSST3 column(150 mm×2.1 mm,1.8μm),the mobile phase was acetonitrile-0.1% trifluoroacetic acid(gradient elution),the flow rate was 0.30 m L/min,the detection wavelength was 254 nm,and the column temperature was 30℃.4-hydroxybenzoic acid was taken as reference,the UPLC chromatograms of 18 batches of herbal pieces were determined.The Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2012.0)saftware was used to evaluate the similarity,determine the common peaks,and carry out cluster analysis and principal component analysis.Results There were 13 common peaks in the UPLC fingerprints of Leonuri Fructus,and the similarity of Leonuri Fructus from different habitats was in the range of 0.801-0.982.Eighteen batches of Leonuri Fructus could be classified into three groups.Peak 4,peak 10 and peak 12 were the important factors to judge the quality of Leonuri Fructus from different habitats.Conclusion The quality of Leonuri Fructus from different habitats is significantly different.The established UPLC fingerprints can provide a reference for the authenticity identification and quality evaluation of Leonuri Fructus.
