超声波辅助乙醇-硫酸铵双水相体系提取绞股蓝总黄酮工艺研究 被引量:6

摘要 本文采用超声波辅助乙醇-硫酸铵双水相体系提取绞股蓝总黄酮,并优化其工艺条件。在单因素试验的基础上结合正交试验,以总黄酮提取量为指标,探讨料液比、乙醇浓度、硫酸铵用量、超声功率和超声温度对绞股蓝总黄酮提取量的影响。结果表明:超声辅助乙醇-硫酸铵双水相提取绞股蓝总黄酮的最优工艺为:料液比1:30(g/mL),乙醇浓度50%,硫酸铵用量0.25 g/mL,超声功率240 W,超声时间40 min,超声温度70℃。在此条件下,总黄酮提取量为15.90 mg/g。超声波辅助乙醇-硫酸铵双水相提取绞股蓝总黄酮的效果较好,是提取绞股蓝中总黄酮的有效方法。 Ultrasonic assisted ethanol-ammonium sulfate aqueous two-phase system was used to extract the total flavonoids from Gynostemma Pentaphyllum.On the basis of single factor test and orthogonal test,the extraction amount of total flavonoids was taken as the index to investigate the influence of the ratio of solid to liquid,the concentration of ethanol,the amount of ammonium sulfate,the ultrasonic power and the ultrasonic temperature on the extraction amount of total flavonoids from Gynostemma Pentaphyllum.The optimal ultrasonic assisted aqueous two-phase extraction process of total flavonoids from Gynostemma Pentaphyllum was as follows:solid-liquid ratio was 1:30(g/mL),ethanol concentration was 50%,ammonium sulfate dosage was 0.25 g/mL,ultrasonic power was 240 W,ultrasonic time was 40 min,ultrasonic temperature was 70℃,under this condition,the extraction amount of total flavonoids was 15.90 mg/g.Ultrasonic assisted ethanol-ammonium sulfate aqueous two-phase extraction of the total flavonoids from Gynostema pentaphylla has a high yield and is an effective method to extract total flavonoids from Gynostema Pentaphylla.
