蒙药蜀葵花DNA条形码分子鉴定技术鉴定与功效组分含量测定 被引量:3

Identification by DNA barcoding molecular identification technique and active components content determination for marshmallow flowers in Mongolian medicine

摘要 目的建立蜀葵花脱氧核糖核酸(DNA)编码技术鉴定及功效组分的含量测定方法。方法采用DNA条形码分子鉴定技术进行聚合酶链式反应(PCR)扩增并测定标记(matK)序列;应用高效液相色谱法(HPLC)测定功效组分槲皮素、木犀草素、山柰素的含量。结果经DNA条形码分子鉴定技术鉴定了11批蜀葵的干燥花药材的matK序列;功效组分含量测定方法中槲皮素、木犀草素、山柰素的平均回收率(n=6)依次为97.75%(RSD=0.33%)、100.04%(RSD=1.55%)、97.05%(RSD=0.86%)。结论DNA条形码分子鉴定技术鉴定方法准确可靠,功效组分含量测定方法专属性强、重复性好,二者结合可作为该药的质量控制方法。 Objective To establish a method for the identification by deoxyribonucleic acid(DNA)coding technology and the active component content determination for marshmallow flowers.Methods Polymerase chain reaction(PCR)was used to amplify and determine the mark(Matk)sequences by DNA barcoding molecular identification technology,and High Performance Liquid Chromatography(HPLC)was applied to determine the content of active components,such as quercetin,luteolin and kaempferide.Results Eleven lots of dried marshmallow flowers(Althaea rosea L.)Cavan were identified for matK sequences by DNA barcoding molecular identification technique;the mean recoveries(n=6)of quercetin,luteolin and kaempferide in the content determination of the active components were 97.75%(RSD=0.33%),100.04%(RSD=1.55%),and 97.05%(RSD=0.86%).Conclusion The DNA barcoding molecular identification method is accurate and reliable,and the active component content determination method is specific and repeatable,so the combination of the two can be used as a quality control method for marshmallow flowers.
