摘要
以香菇为主要原料,采用连续动态逆流水提醇沉法提取香菇多糖并以热水回流提取法作为对照,对比动态逆流提取对香菇多糖提取得率和抗氧化能力的影响。采用苯酚硫酸法测定多糖含量,检测香菇多糖清除DPPH·、·OH、O^(2-)·自由基能力以及Fe离子还原力。结果表明,连续动态逆流提取浓度平衡后20 min取样,香菇多糖得率最高(8.43%),是热水回流提取法2.63倍。连续动态逆流提取法比热水回流法提取的香菇多糖对羟自由基、超氧自由基、DPPH自由基的清除能力更强,接近维生素C的抗氧化能力。当香菇多糖质量浓度为1.5 mg/mL时,连续动态逆流提取法提取的香菇多糖对O^(2-)·、·OH和DPPH·清除率分别为71.35%、78.36%和70.36%,比热水回流提取的香菇多糖抗氧化能力提高了38.75%,52.36%和36.82%。
With Lentinus edodes as the main raw material,polysaccharide is extracted from Lentinus edodes by continuous dynamic countercurrent water extraction with hot water reflux extraction as control.The extraction rate and antioxidant activity of Lentinus edodes polysaccharide are compared.The content of polysaccharide is determined by phenol-sulfuric acid method.The scavenging ability of Lentinus edodes polysaccharide on·OH,O^(2-)·,DPPH·free radicals and the reducing power of Fe3+are detected.The results show that the yield of Lentinus edodes polysaccharide is the highest(8.43%)20 min after the concentration achieves a balance by the continuous dynamic countercurrent extraction,which is 2.63 times that of hot water reflux extraction.Lentinus edodes polysaccharide extracted by continuous dynamic countercurrent extraction has stronger scavenging ability on·OH,O^(2-)·and DPPH·free radicals,and is close to the antioxidant capacity of vitamin C.When the concentration of Lentinus edodes polysaccharide is 1.5 mg/mL,the scavenging rate of Lentinus edodes polysaccharide extracted by continuous dynamic countercurrent extraction for·OH,O^(2-)·and DPPH·is 71.35%,78.36%and 70.36%,respectively,and its antioxidant activity is increased by 38.75%,52.36%and 36.82%,respectively compared to Lentinus edodes polysaccharide extracted by hot water reflux extraction.
