生物基质中雌激素的液相色谱-串联质谱定量分析方法研究进展 被引量:2

摘要 雌激素是一类重要的内源性甾体类激素,具有广泛的生物活性。内源性雌激素及其代谢物含量的测定对生理和病理学诊断具有极其重要的意义。然而,雌激素在生物基质中浓度通常极低,复杂基质干扰大,难以准确定量检测。液相色谱-串联质谱法(LC-MS/MS)具有高灵敏度、高准确性以及高通量的优势,已经成为生物基质中雌激素检测的主要技术平台。本文主要从样品前处理、衍生化方法、色谱-质谱的仪器与方法,以及不同类型的生物基质中雌激素的检测结果四个方面,综述了近五年来基于LC-MS/MS联用技术应用于生物基质中雌激素检测的最新研究进展以及趋势,讨论了目前生物基质中雌激素的定量分析存在的问题,并对其前景进行了展望。 As an important endogenous steroid hormone,estrogen has a wide range of biological activities.The determination of endogenous estrogen and its metabolites is crucial to physiological and pathological diagnosis.However,the concentration of estrogen in the biological matrix is usually very low and the matrix interference is complicated.Therefore,it is difficult to accurately analyze the estrogen content in biological matrix.Liquid chromatography-tandem mass spectrometry has the advantages of high sensitivity,high accuracy and high throughput,and has become one of the main technology platforms for estrogen analysis in biological matrix.This review summarizes the latest research progress and trends on the detection of estrogen in biological matrix by liquid chromatography-tandem mass spectrometry technology in the past five years,including sample types,pretreatment,derivatization methods,and liquid chromatography-mass spectrometry analysis etc.It also discusses existing problems and future prospective with respect to estrogen quantification.
