基因编辑技术及其在柞蚕基础研究中的应用前景

摘要 基因编辑技术是功能基因研究和生物遗传改良的重要手段。本文以第三代人工核酸内切酶编辑技术成簇的规律间隔的短回文重复序列系统(CRISPR/Cas)为主,以鳞翅目模式生物——家蚕为例,简要介绍了基因编辑技术的原理及研究成果,总结了开展柞蚕基因编辑可能遇到的问题及相应解决策略,展望了基因编辑技术在柞蚕基础研究中的应用前景,以期为柞蚕后基因组时代的基因功能分析、突变模型建立、品种遗传改良等研究提供参考。 Gene editing is an important means of functional gene research and biological genetic improvement.In this paper,the principle and research results of gene editing technology were briefly introduced by primarily using the third generation of artificial endonuclease editing technology,CRISPR/Cas system,and taking the Lepidopteran model organism,Bombyx mori,as an example.It summarized possible problems and corresponding solutions when performing gene editing in tussah,and looked forward to the application prospect of gene editing in tussah in order to provide ideas for the studies of gene function analysis,mutation model establishment and genetic improvement of varieties in tussah post-genomic era.
