摘要
目的:寻找糖尿病气阴两虚证潜在生物标志物及相关的代谢通路,探讨其内在的生物物质基础。方法:运用超高效液相色谱与串联四级杆飞行时间质谱仪(Ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry,UPLC/Q-TOF-MS)联用技术检测模型组和空白组生物样本血清的代谢轨迹,采用主成分分析(principal component avalysis,PCA)和偏最小二乘法判别分析(partial least squares discrimination analysis,PLS-DA)寻找各组代谢物差异,筛选潜在内源性标志物。结果:发现与疾病相关的代谢物变化涉及碳水化合物代谢8条,脂质代谢11条,蛋白质代谢10条,氧化应激损伤代谢7条,能量代谢5条。结论:糖尿病气阴两虚证生物模型存在碳水化合物代谢、脂质代谢、蛋白质代谢、氧化应激、三羧酸循环及生物能量代谢紊乱,代谢组学研究方法有助于揭示中医证候的微观物质基础。
Objective:To search for potential biomarkers and related metabolic pathways in diabetes mellitus(DM)with deficiency of both qi and yin syndrome,and to explore their intrinsic biological basis.Methods:Ultra performance liquid chromatography coupled with tandem quadrupole time-of-flight mass spectrometry(UPLC/Q-TOF-MS)was used to detect the metabolic trajectory of serum samples from the model group and blank group.Principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were used to find the differences of metabolites in each group and screen potential endogenous markers.Results:It was found that the changes of metabolites related to diseases involved 8 carbohydrate metabolism,11 lipid metabolism,10 protein metabolism,7 oxidative stress injury metabolism and 5 energy metabolism.Conclusion:Metabolites of carbohydrate metabolism,lipid metabolism,protein metabolism,oxidative stress,three carboxylic acid cycle and bioenergy metabolism are found in the biological models of DM with deficiency of both qi and yin syndrome in two cases.Metabonomics research helps to reveal the microscopic material foundation of TCM syndromes.
