摘要
目的检测网钙结合蛋白2(reticulocalbin2,RCN2)在甲状腺乳头状癌(papillary thyroid carcinoma,PTC)组织中的表达情况,探讨RCN2蛋白异常表达在该病中的临床意义。方法应用免疫组织化学法和蛋白质印迹法检测65例甲状腺乳头状癌组织及其癌旁组织中RCN2蛋白的异常表达情况,分析其在该病中的临床意义。结果65例甲状腺乳头状癌患者中,免疫组化法检测RCN2蛋白在癌组织中的表达率为69.23%(45/65),明显高于癌旁正常组织18.46%(12/65),差异有统计学意义(χ^(2)=34.023,P<0.001),蛋白质印迹法结果显示RCN2蛋白在癌组织中的相对表达量(5.17±0.60)明显高于癌旁组织(3.37±0.57),差异有统计学意义(t=27.665,P<0.001)。甲状腺乳头状癌组织中RCN2蛋白高表达与患者年龄、肿瘤直径、肿瘤数量、TNM分期、淋巴结转移均有关(P<0.05)。结论RCN2蛋白在甲状腺乳头状癌组织中呈高表达,且均与肿瘤恶性进展关系密切,为甲状腺乳头状癌临床治疗提供潜在靶点。
Objective To detect the expression of calcium binding protein 2(reticulocalbin2,RCN2)in tissues of papillary thyroid carcinoma(papillary thyroid carcinoma,PTC)and explore the clinical significance of abnormal expression of RCN2 protein in the disease.Methods Immunohistochemistry and western blotting methods were used to detect the abnormal expression of RCN2 protein in 65 cases of papillary thyroid carcinoma and its adjacent tissues to analyze its clinical significance in the disease.Results Among 65 patients with papillary thyroid carcinoma,the expressive rate of RCN2 protein was 69.23%(45/65)through immunohistochemical method,significantly higher than 18.46%(12/65)of normal tissues adjacent to cancer.The difference was statistically significant(χ^(2)=34.023,P<0.001).The expression of RCN2 in cancer tissues(5.17±0.60)was significantly higher than para-carcinoma tissue(3.37±0.57).The difference was statistically significant(t=27.665,P<0.001).The high expression of RCN2 protein in papillary thyroid carcinoma was related to age,tumor diameter,tumor number,TNM stage and lymph node metastasis(P<0.05).Conclusion RCN2 protein is highly expressed in papillary thyroid carcinoma and is closely related to malignant progression of thyroid carcinoma,which provides a potential target for clinical treatment of papillary thyroid carcinoma.
