摘要 目的利用生物信息学方法探究胶原蛋白VIα1[Collagenα-1(VI)gene,COL6 A1]基因表达量与肾透明细胞癌患者临床病理特征及生存预后之间的关系。方法所有数据均使用R软件进行分析,利用R语言的生存包构建生存预后模型,利用基因集富集分析预测肾透明细胞癌中与COL6A1相关的信号通路。结果肾透明细胞癌组织中COL6A1表达显著高于正常肾组织。COL6A1表达量与肾透明细胞癌患者的年龄、病理分级、T分期、N分期、临床分期显著相关。COL6A1高表达组的患者预后较差且COL6A1高表达是肾透明细胞癌患者不良预后的独立风险因素。C指数、校准曲线和ROC曲线均证明3年和5年总生存率预测模型与实际基本相符。基因集富集分析显示许多关键通路被富集到COL6A1高表达表型。结论本研究结果表明COL6A1可能是肾透明细胞癌不良预后的潜在生物标志,且COL6A1可能通过与许多关键信号通路相互作用参与肾透明细胞癌的进展。 Objective Bioinformatics method was used to investigate the relationship between COL6A1 gene expression and clinicopathological characteristics and survival prognosis of patients with clear cell renal carcinoma.Methods All data were analyzed by R software.A survival prognosis model was constructed by using survival package of R language.Gene Set Enrichment Analysis was used to analyze and predict the signaling pathway related to COL6A1 in clear cell renal carcinoma.Results The expression of COL6A1 in clear cell renal carcinoma tissues was significantly higher than that in normal renal tissues.The expression level of COL6A1 was significantly correlated with age,pathological grade,T stage,N stage and clinical stage of patients with clear cell renal carcinoma.Patients with high COL6A1 expression had a poor prognosis and high COL6A1 expression was an independent risk factor for poor prognosis in patients with clear cell renal carcinoma.C index,calibration curve and ROC curve all proved that the prediction model of 3-year and 5-year overall survival rate was basically consistent with the reality.Gene enrichment analysis showed that many key pathways were enriched into the highly expressed COL6A1 phenotype.Conclusion The results of this study suggest that COL6A1 may be a potential biomarker for poor prognosis of renal clear cell carcinoma,and that COL6A1 may be involved in the progression of renal clear cell carcinoma through interactions with many key signaling pathways.
