摘要
目的检测微小RNA-335-5p(miR-335-5p)、肿瘤坏死因子受体超家族成员9(CD137)在急性心肌梗死(AMI)患者血清中的表达情况,探讨两者对AMI的影响及与预后的关系。方法本研究选取2015年3月至2017年6月于河南省第二人民医院心内科收治的已确诊AMI患者85例为AMI组,另选取同时期入院健康体检者73例为对照组。采用实时荧光定量PCR(qRT-PCR)检测血清中miR-335-5p的表达情况;ELISA法检测血清中CD137、心肌肌钙蛋白I(cTnI)和肌红蛋白(MyO)的表达情况,采用Pearson法分析CD137、miR-335-5p与cTnI、MyO间的相关性,探讨两者表达水平与预后的关系。结果与对照组相比,AMI组患者的CD137、cTnI和MyO表达水平显著增加,而miR-335-5p表达水平则显著降低(P<0.05);随着发病时间的延长cTnI、MyO、CD137表达水平在4~8 h内持续增加,而miR-335-5p表达水平则持续降低。Pearson法分析发现,AMI患者血清中CD137与cTnI、MyO表达均呈明显正相关关系(P<0.05),而miR-335-5p与cTnI、MyO表达均呈明显负相关关系(P<0.05);此外,CD137低表达与高表达组相比预后生存率更高,miR-335-5p高表达与低表达组相比患者预后生存率更高。结论AMI患者血清中CD137表达水平显著增加,miR-335-5p表达水平则显著降低,其表达量可作为检测AMI发病的重要参照指标之一,并可用于判断病情严重程度及预后。
Objective The expression of microrna-335-5p(miR-335-5p),tumor necrosis factor receptor superfamily member 9(CD137)in the serum of patients with acute myocardial infarction(AMI)was detected to explore the impact of the two on AMI and the relationship with prognosis.Methods In this study,85 AMI patients who had been diagnosed and treated at the Department of Cardiology of the Second People's Hospital of Henan from March 2015 to June 2017 were selected as the AMI group,and 73 healthy individuals as the control group.Real-time PCR(QRT PCR)was used to determine the expression of mir-335-5p in the child's serum,ELISA was used to determine the expression of CD137,cardiac troponin I(cTnI)and myoglobin(Myo),Pearson's test was used to determining the correlation between the expression levels of CD137,mir-335-5p and cTnI,Myo,and to investigate the relationship between the expression levels and prognosis.Results Compared with those in the control group,the expression levels of CD137,cTnI and MyO in AMI group were significantly higher,while the expression level of miR-335-5p was significantly lower(P<0.05);with the prolongation of the onset time,the expression levels of cTnI,MyO and CD137 continued to increase within 4-8 h,however,the expression level of miR-335-5p continued to decrease.Pearson analysis showed that CD137 was significantly positively correlated with cTnI and MyO(P<0.05),while miR-335-5p was significantly negatively correlated with cTnI and MyO(P<0.05);in addition,the survival rate of CD137 low expression group was higher than that of high expression group,and the prognosis survival rate of patients with high expression of miR-335-5p was higher than that of low expression group.Conclusions CD137 expression was significantly increased,while mir-335-5p was decreased in the serum of patients with AMI.It can be used as one of the important reference indicators for detecting the onset of AMI as well as to determine the severity and prognosis of the disease.
