酶解豆清液制备抗氧化肽 被引量:4

Preparation of Antioxidant Peptides by Enzymatic Hydrolysis of Soybean Wastewater

摘要 以豆清液为原料,用胃蛋白酶酶解豆清液制备豆清多肽,以多肽产率为指标,探讨酶解时间、酶与底物浓度比、初始p H和酶解温度对多肽产率的影响;用Box-Behnken中心组合原理建立数学模型,通过响应面优化提取工艺参数;测定ABTS自由基清除率和DPPH自由基清除率。结果表明,酶解最优条件为时间7 h、酶与底物浓度比0.8g/m L、初始p H 3.5、酶解温度31℃,在此条件下多肽产率为40.08%;在抗氧化试验中,在0~0.05 g/mL浓度范围内,豆清多肽0.05 g/mL时对ABTS自由基清除率最大,达到100%,对DPPH自由基清除率最大,达到98.8%。结果表明,胃蛋白酶酶解制备的豆清多肽具有较好的抗氧化性,是优质抗氧化肽的良好来源。通过胃蛋白酶酶解可有效提高豆清多肽产率,旨在为豆清液的再利用提供理论依据。 Using soybean wastewater as the raw material,preparation of soybean peptide by enzymolysis with pepsin,and the peptide yield was used as an indicator to explore the effects of enzymolysis time,enzyme dosage,initial pH and enzymolysis temperature on the peptide yield.The Box-Behnken central combination principle was used to establish a mathematical model,and extract process parameters were optimized through response surface design.The ABTS free radical scavenging rate and DPPH free radical scavenging rate were measured.The results showed that the optimal condition for enzymatic hydrolysis was 7 h,the enzyme dosage 0.8 g/mL,the initial pH 3.5,and the enzymatic hydrolysis temperature 31℃.Under this condition,the peptide yield was 40.08%.In the antioxidant experiment,the scavenging rate of ABTS free radicals at 0.05 g/mL reached the maximum 100%,and the scavenging rate of DPPH free radicals reached the maximum 98.8%.The results indicated that the soybean peptide prepared by pepsin hydrolysis had better antioxidant properties and was a good source of high-quality antioxidant peptides.Pepsin digestion could effectively increase the yield of soybean peptide,to provide a theoretical basis for the reuse of soybean wastewater.
