簕菜根中质量标志物的初步预测及分析

Preliminary Prediction and Analysis of Quality Markers in Acanthopanax trifoliatus(L.)Merr.

摘要 基于刘昌孝院士提出的质量标志物(Q-marker)理念,预测簕菜根的质量标志物并评价簕菜根的质量。基于簕菜根亲缘关系与生源途径、化学成分、药理作用的关系,预测簕菜根的Q-marker;建立高效液相色谱法同时测定其质量标志物成分。结果表明,预测酚酸类成分是簕菜根的主要活性物质之一,可作为Q-marker。新绿原酸、绿原酸、异绿原酸B、异绿原酸A、异绿原酸C分别在0.019~0.380,0.390~7.800,0.033~0.660,0.200~4.000和0.060~1.200mg/m L范围内线性关系良好,其平均加样回收率分别为100%,105%,98%,103%和104%。10个产地的簕菜根中均能检测出这5个成分。以酚酸类成分作为簕菜根的Q-marker科学合理,建立的簕菜根中酚酸类成分的HPLC测定方法可用于簕菜根质量的控制。 Based on the concept of Q-marker proposed by academician LIU Changxiao,to predict the Q-marker of Acanthopanax trifoliatus(L.)Merr and evaluate the quality of Acanthopanax trifoliatus(L.)Merr,the Q-marker was preliminary predicted,based on the relationship between genetic relationship and biosynthesis pathway,chemical composition and pharmacological activity;The Q-marker components were determined by HPLC.The results showed that phenolic acid components were predicted to be one of the main active substances of Acanthopanax trifoliatus(L.)Merr.,which could be used as the main selection of Q-marker.Neochlorogenic acid,chlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C had good interlineal relations at 0.019-0.380,0.390-7.800,0.033-0.660,0.200-4.000 and 0.060-1.200 mg/mL,respectively.The average recovery rates were 100%,105%,98%,103%and 104%,respectively.These 5 components can be detected in Acanthopanax trifoliatus(L.)Merr.from 10 producing areas.Taking phenolic acid components as Q-marker of Acanthopanax trifoliatus(L.)Merr.is scientific and reasonable.HPLC method for determination of phenolic acid components in Acanthopanax trifoliatus(L.)Merr.can be used for quality control of its medicinal materials.
