干旱胁迫对三七生理指标的影响及转录组分析 被引量:6

摘要 为揭示干旱胁迫下三七的生理特性和基因转录组变化,通过干旱条件和正常浇水控制研究干旱对于三七的影响。实验设置自然干旱和正常浇水(对照)处理,分别测定三七叶片的叶绿素含量、可溶性蛋白含量、POD活性、SOD活性、CAT活性和MDA活性,并从三七叶片中提取RNA进行转录组测序分析。结果显示,干旱胁迫下三七MDA变化非常明显,与对照相比,植株受到干旱后的MDA活性升高了88.41%,相反CAT活性受干旱影响的效果不明显。转录组分析结果表明,与对照相比,干旱处理样品上调基因431个,下调基因151个,对差异基因进行GO注释和功能富集性分析,有160个上调基因和53个下调基因共213个差异基因注释到数据库中。KEGG分析结果表明,有122个DEGs在37个代谢通路上。ABA合成代谢的关键酶,9-顺式-环氧类胡萝卜素双加氧酶被显著富集,说明干旱条件下,三七可能通过ABA相关合成酶的增加调控干旱应答。 To reveal the physiological characteristics and gene transcriptome changes of Panax notoginseng under drought stress, we studied the effects of drought on P. notoginseng through drought conditions andnormal watering(control). The chlorophyll content, soluble protein content, POD activity, SOD activity, CATactivity and MDA activity of P. notoginseng leaves were measured respectively, and RNA was extracted from P.notoginseng leaves for transcriptome sequencing analysis. The results show that the change of MDA in P.notoginseng is very obvious under drought stress. Compared with the control, the MDA activity of the plantincreases by 88.41% after drought, on the contrary, the effect of CAT activity affected by drought is notobvious. The results of transcriptome analysis show that there are 431 up-regulated genes and 151 down-regulated genes in the drought treated samples compared with the control. GO annotation and functionalenrichment analysis were carried out on the differential genes. There are 160 up-regulated genes and 53 down-regulated genes with 213 differential gene annotations in the database. KEGG analysis shows that there are 122 DEGs in 37 metabolic pathways. 9-cis-cyclocarotenoid-dioxygenase, the key enzyme of ABA anabolism, is significantly enriched, indicating that P. notoginseng may regulate drought response through the increase ofABA related synthase.
