摘要
目的研究急性心肌梗死(AMI)患者行冠状动脉(冠脉)介入治疗时发生缺血再灌注后体内循环微粒(MPs)数量及质量的改变。方法选取AMI行冠脉介入治疗时发生缺血再灌注的患者(30例)为实验组,对照组为稳定型心绞痛并行冠脉介入治疗的患者(30例),于冠脉介入治疗结束时抽取动脉血并离心提取MPs,检测两组MPs浓度,通过流式细胞仪检测MPs来源,最后用提取的MPs孵育离体大鼠胸主动脉检测一氧化氮(NO)的含量。结果与对照组MPs比较,实验组MPs含量明显升高,流式细胞仪检测发现实验组MPs主要为血小板来源,功能学检测显示实验组明显抑制离体大鼠胸主动脉NO的产生。结论缺血再灌注可能通过增加冠脉内血小板和内皮来源的MPs影响冠脉血管的舒张和血液供应。
Objective To study the changes in quantity and quality of microparticles(MPs)after ischemia reperfusion in patients with acute myocardial infarction(AMI)during coronary intervention therapy.Methods AMI patients with ischemia reperfusion during coronary intervention therapy were chosen into test group(n=30),and patients with stable angina pectoris(SAP)undergone coronary intervention therapy were chosen into control group(n=30).The samples of artery blood were centrifuged for extracting MPs after coronary intervention therapy,and concentration of MPs was detected in 2 groups.The origin of MPs was detected by using flow cytometer,and finally the content of nitric oxide(NO)was detected in vitro in rat thoracic aorta incubated with extracted MPs.Results The content of MPs increases significantly in test group compared with control group.The results of flow cytometer detection showed that MPs were mainly platelet-derived MPs.Functional detection showed that NO production was significantly inhibited in vitro in rat thoracic aorta test group.Conclusion Ischemia reperfusion may influence coronary vasodilatation and blood supply through increasing platelet-derived MPs and endothelial-derived MPs in coronary artery.
