摘要
目的观察中药芪卫颗粒对体外高糖诱导的小鼠肾小球足细胞转分化的作用及对细胞miRNA-155表达的影响,探讨其保护足细胞的可能机制。方法以小鼠肾足细胞株作为研究对象,采用高糖刺激构建细胞转分化模型,分为正常组(空白血清培养)、高糖组(30 mmol/L葡萄糖+空白血清培养)、芪卫颗粒组(30 mmol/L葡萄糖+含药血清培养)、miRNA-155基因敲减组(30 mmol/L葡萄糖+miRNA-155抑制剂)4个组,48小时后,收集各组细胞并进行相关检测。CCK-8法检测各组细胞活力,实时荧光定量聚合酶链式反应及蛋白质印迹法检测各组细胞转分化相关指标nephrin和α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)的表达水平,各组细胞miRNA-155的表达水平采用实时荧光定量聚合酶链式反应检测。结果相比较于正常组,高糖组细胞活力下降(P<0.05),转分化相关指标nephrin的基因和蛋白表达均减少(P<0.05),α-SMA的基因和蛋白表达均增加(P<0.05),miRNA-155表达水平明显升高(P<0.05);而与高糖组相比,芪卫颗粒组和miRNA-155基因敲减组细胞活力有所升高(P<0.05),nephrin的基因和蛋白表达均明显上调(P<0.05),α-SMA的基因和蛋白表达均有所下调(P<0.05),且芪卫颗粒组miRNA-155表达水平较高糖组明显减少(P<0.05)。结论芪卫颗粒能通过抑制miRNA-155的水平来减轻高糖诱导的足细胞转分化。
Objective To observe effects of Qiweikeli on glomerular podocytes epithelial-mesenchymal transition(EMT)induced by high glucose and miRNA-155.Methods Mouse podocytes were randomly divided into 4 group:normal group(blank serum),model group(blank serum with 30 mmol/L glucose),Qiweikeli group(medicine serum with 30 mmol/L glucose)and miRNA-155 knockout group(blank serum with 30 mmol/L glucose in the absence of miRNA-155).The cells of each group were collected and detected after 48 hours.Podocytes viability was determined by CCK-8 assay.Expression of nephrin,α-smooth muscle actin(α-SMA)and miRNA-155 were assessed by real-time PCR or western blot.Results High glucose could trigger podocyte EMT,with the presentation of decreased nephrin mRNA and protein expression,and increasedα-SMA mRNA and protein expression as well as reduced podocytes viability and increased miRNA-155 levels.Compared with model group,both Qiweikeli group and miRNA-155 knockout group could improve high glucose-induced podocyte EMT through increasing nephrin mRNA and protein expression and reducingα-SMA mRNA and protein expression,including enhanced podocytes viability and reduced miRNA-155 levels.Conclusion Qiweikeli could alleviate glomerular podocyte EMT induced by high glucose through inhibiting the expression of miRNA-155.
