趋化因子受体表达与结直肠癌淋巴结转移的关系研究

Relationship between expression of chemokine receptors and lymph node metastasis in colorectal cancer

目的探讨趋化因子受体(CCR)表达与结直肠癌(CRC)淋巴结转移的关系。方法选取2018年1月至2019年12月在衢州市人民医院行根治手术的新发CRC患者100例为研究对象,其中淋巴结转移阳性37例,阴性63例。采用RT-qPCR法检测CRC组织中相关分子标志物mRNA表达,免疫荧光法检测淋巴结中相关分子标志物蛋白表达。结果淋巴结转移阳性患者CRC组织中CCR2、CCR4 mRNA相对表达量均明显高于淋巴结转移阴性患者,差异均有统计学意义(均P<0.05);淋巴结转移阳性与阴性患者CRC组织中CCR7、转化生长因子-β(TGF-β)R1、p53、CD3e、CD8a、IL-2RA mRNA相对表达量比较,差异均无统计学意义(均P>0.05)。在倒置荧光显微镜下,正常淋巴结中CCR2、CCR4呈低表达,但在转移性淋巴结中表达显著增强。结论CCR2、CCR4表达与CRC淋巴结转移有关。

Objective To investigate the relationship between the expression of chemokine receptors(CCR)and lymph node metastasis in colorectal cancer(CRC).Methods From January 2018 to December 2019,a total of 100 newly diagnosed CRC patients in Quzhou People's Hospital were enrolled,including 37 cases with positive lymph node metastasis and 63 cases with negative metastasis.The mRNA and protein expression of relative molecular markers in colorectal cancer tissues and in lymph nodes were detected with RT-qPCR and immunofluorescence,respectively.Results The expressions of CCR2 and CCR4 mRNA in CRC tissues from patients with positive lymph node metastasis were significantly higher than those of patients with negative lymph node metastasis(all P<0.05).There was no significant difference in the expression of CCR7,transforming growth factor-β(TGF-β)R1,p53,CD3e,CD8a,IL-2RA mRNA in CRC tissues between patients with positive and negative lymph node metastasis(all P>0.05).Immunofluorescence showed that the expression of CCR2 and CCR4 was low in normal lymph nodes,whereas the expression was significantly increased in metastatic lymph nodes.Conclusion The expression of CCR2 and CCR4 may be related to CRC lymph node metastasis in colorectal cancer.
