糖交联胶原-羟基磷灰石复合支架材料对人成骨样细胞的黏附影响

摘要 目的在糖交联胶原-羟基磷灰石颗粒支架材料表面接种人成骨样SAOS-2细胞,评价其对SAOS-2细胞活性、黏附的影响。方法采用扫描电子显微镜观测材料表面和剖面内细胞生长形态,通过细胞示踪、免疫荧光染色后激光共聚焦显微镜观测细胞活性和在材料表面/内部的黏附。结果人成骨样SAOS-2细胞可在支架材料表面黏附,伸展伪足迁移入支架材料内部黏附生长,支架材料上的SAOS-2表达细胞黏附因子Integrin-α2β1。结论人成骨样SAOS-2细胞可在糖交联胶原-羟基磷灰石支架材料表面生长并长入材料海绵状孔隙内,该材料表现出良好的生物相容性,人成骨样SAOS-2细胞与I型胶原纤维基质黏附界面可检测到整合素Integrin-α2β1表达。 Objective To inoculate human osteoblast-like cells on the surface of Hydroxyapatite reinforced sugar cross-linked collagen scaffolds and evaluate cell activity and adhesion.Methods Cell-scaffolds complex were co-cultured for 3 weeks.The cell morphology on the surface and interior space of the material was observed by scanning electron microscopy.The cell activity and adhesion were observed by laser confocal microscopy after cell tracing and immunofluorescence staining.Results Human osteoblast-like cells adhered to the surface of scaffolds,then penetrated into the interior of the sponge-like scaffold and expressing Integrin-α2β1.Conclusions SAOS-2 seeded on the sugar cross-linked collagen scaffolds together with hydroxyapatite showed a proper cellular attachment,penetration and migration.The expression of Integrin-α2β1 was detected in SAOS-2.Sugar cross-linked collagen scaffolds together with hydroxyapatite may induce osteogenic differentiation of SAOS-2,possibly through activating integrins.
