摘要
目的运用代谢组学的方法研究抽动障碍(tic disorder,TD)患儿血清代谢产物差异,总结TD患儿血清相关生物标志物特点,探析TD的生物学实质,分析其代谢通路及生物学意义,为TD的诊断及进一步研究提供思路。方法选取2019年10月—2020年3月期间于北京中医药大学东方医院儿科门诊确诊为TD的患儿30例(TD组),同时选取同一时期体检健康儿童30例设为正常组。采用液-质联用技术(LC-MC)、气-质联用技术(GC-MC)和化学计量学分析研究技术,检测TD患儿血清代谢物变化,筛选出TD代谢生物标记物。结果筛选出与TD相关的生物标记物为甘油三酯、L-苯丙氨酸-L-脯氨酸、乳糖神经酰胺、白三烯、2-甲基-3-酮戊酸、L-胱氨酸、硫酸脱氢异雄酮、聚乙烯等50余种,主要涉及甘油磷脂代谢、亚油酸代谢、花生四烯酸代谢等代谢通路。结论所筛选的生物标记物及其代谢通路,可作为TD患儿独特的代谢图谱,成为TD的诊断依据及进一步研究靶点。
Objective To use metabonomics methods to study the differences in serum metabolites of children with tic disorder(TD),summarize the characteristics of serum-related biomarkers in children with TD,explore the biological essence of TD,and analyze its metabolic pathways and biology Significance,to provide ideas for the diagnosis and further research of TD.Methods Select 30 children with TD who were clearly diagnosed as TD in the Pediatric Clinic of Dongfang Hospital of Beijing University of Chinese Medicine from October 2019 to March 2020.At the same time,30 healthy children with physical examination during the same period were selected as the normal group.Using liquid-mass spectrometry(LC-MC),gas-mass spectrometry(GC-MC)and chemometric analysis and research techniques,the changes in serum metabolites of children with TD were detected,and TD metabolic biomarkers were screened out.Results The biomarkers related to TD were selected as triglyceride,L-phenylalanine-L-proline,lactose ceramide,leukotriene,2-methyl-3-ketovaleric acid.There are more than 50 kinds of cystine,dehydroepiandrosterone sulfate,polyethylene,etc.,mainly related to metabolic pathways such as glycerophospholipid metabolism,linoleic acid metabolism,and arachidonic acid metabolism.Conclusion The selected biomarkers and their metabolic pathways can be used as a unique metabolic profile for children with TD,and become a diagnostic basis for TD and a target for further research.
