长链非编码RNA PEG10在结直肠癌中表达的临床意义和生物学功能 被引量:1

摘要 目的探讨长链非编码RNA(long non-coding RNA,lncRNA)父系表达遗传印记基因10(paternally expressed 10,PEG10)在人结直肠癌(colorectal cancer,CRC)中的表达、临床意义及生物学作用。方法利用实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,qRT-PCR)的方法检测66例CRC肿瘤组织及正常癌旁对照组织中lncRNA PEG10 mRNA的表达水平,分析lncRNA PEG10的表达与CRC患者临床病理资料及预后之间的相关性。在体外,利用CCK-8实验、Transwell迁移和侵袭实验检测lncRNA PEG10的表达对CRC细胞增殖、迁移和侵袭能力的影响。结果与癌旁正常对照组织相比,lncRNA PEG10在CRC肿瘤组织中的表达显著升高(P<0.01),并且与CRC患者的肿瘤浸润深度(P=0.017)、淋巴结转移情况(P=0.009)及TNM分期(P<0.001)显著相关。此外,lncRNA PEG10高表达是CRC患者预后的独立危险因素,与CRC患者总生存期(P=0.0037)和无进展生存期(P=0.0019)的减少密切相关。CCK-8细胞活性实验表明,lncRNA PEG10表达干扰能够抑制CRC细胞系HCT-116和DLD-1的增殖能力(均P<0.01);Transwell细胞侵袭实验表明,lncRNA PEG10表达干扰能够抑制CRC细胞系HCT-116和DLD-1的浸润能力(P<0.05或P<0.01);Transwell细胞迁移实验表明,lncRNA PEG10表达干扰能够抑制CRC细胞系HCT-116和DLD-1的转移能力(P<0.05或P<0.01)。结论lncRNA PEG10与CRC患者预后不良密切相关,并且促进CRC细胞的增殖、浸润和转移。lncRNA PEG10靶向治疗或许能够为CRC患者预后的改善带来巨大的益处。 Objective To explore the expression,clinical significance and biological functions of long non-coding RNA(lncRNA)paternally expressed 10(PEG10)in human colorectal cancers(CRC).Methods Expression of lncRNA PEG10 was detected by quantitative real-time polymerase chain reaction(qRT-PCR)in 66 cases of CRC tissues and corresponding normal tissues.The correlations of lncRNA PEG10 expression with the clinicopathological characteristics and the prognosis of CRC patients were analyzed.In vitro experiments,cell counting kit-8 assay and Transwell assay were used to evaluate the effects of lncRNA PEG10 on the proliferation,migration and invasion abilities of CRC cells.Results LncRNA PEG10 was significantly upregulated in CRC tissues compared with the corresponding normal tissues(P<0.01),and the expression of lncRNA PEG10 was closely correlated with tumor stage(P=0.017),lymph node metastasis(P=0.009)and tumor-node-metastasis(TNM)stage(P<0.001).In addition,high expression of lncRNA PEG10 was an independent unfavorable prognostic factor,and it was closely associated with overall survival(P=0.0037)and progression-free survival(P=0.0019)in CRC patients.Cell counting kit-8 assay indicated that lncRNA PEG10 inhibition suppressed the proliferative abilities of HCT-116 cells and DLD-1 cells(both P<0.01).Transwell invasion assay indicated that lncRNA PEG10 inhibition suppressed the invasive abilities of HCT-116 cells and DLD-1 CRC cells(P<0.05 and 0.01,respectively).Transwell migration assay indicated that lncRNA PEG10 inhibition suppressed the metastatic abilities of HCT-116 cells and DLD-1 CRC cells(P<0.05 and 0.01,respectively).Conclusion LncRNA PEG10 is closely related with the poor prognosis of CRC patients and can enhance the malignant properties of proliferation,invasion and metastasis in CRC cells.Molecular therapy targeting lncRNA PEG10 might bring significant benefits to the clinical outcome of CRC.
