紫外、氦氖激光等复合诱变产果胶酶细菌ZH_1的研究 被引量:15

摘要 ZH1 菌株是从自然界分离筛选而来的 ,该菌株产果胶酶的最适 pH为 7 0 ;最适温度3 3℃ ;培养 3 6h达产酶高峰 ,酶活力为 73 5 μ/mL ,将ZH1 作为出发菌株 ,经紫外线诱变、硫酸二乙酯诱变 ,亚硝基胍和紫外线复合诱变及氦氖激光等多次反复诱变 ,选育得到一株产果胶酶性稳定且酶活明显提高的突变株ZHg ,其酶活为 3 0 1m/mL ,比出发菌株ZH1 产果胶酶能力提高 3 Strain ZH 1 was segregated and screened from nature.It was facultative aerobe,Gram positive Bacillus Sp.The optimum condition of producing pectinase strain ZH 1 was pH7.0 and temperature33℃.The production of pectinase reached maximum at 36 hour.Enzyme activity of strain ZH 1 was 73.5μ/lm.Strain ZH 1 was done original strain.Complex mutagenests with UV,DES,NTG and UV,many times repeated mutagenests with He-Ne laser,the mutant ZHg was obtained with the stability and obvious improcement of enzyme activity.Its pectinase activity was 301μ/ml,Which was 3.1 times higher than that of the original strain ZH 1.
