脂肪基质血管组分对膝骨关节炎模型大鼠疼痛及软骨修复的研究

摘要 目的观察脂肪基质血管组分(SVF)对膝骨关节炎(KOA)模型大鼠疼痛和软骨修复的作用。方法取大鼠双侧腹股沟脂肪组织制备SVF,采用碘乙酸法构建KOA模型。将25只大鼠随机分为正常组、模型组、SVF低浓度组、SVF中浓度组、SVF高浓度组,每组各5只。采用低(1.6×10^(4)个/mL)、中(3.2×10^(4)个/mL)、高(6.4×10^(4)个/mL)浓度的SVF对大鼠双侧膝骨关节腔注射治疗,每周注射1次,于模型复制后第2周和4周时评价大鼠疼痛指标。注射4周后,取各组大鼠膝关节进行组织病理学观察和OARSI评分。结果模型复制后第2周,与正常组相比,模型组和SVF低、中、高浓度组压痛阈值均降低(P<0.01);模型复制后第4周,模型组压痛阈值低于正常组(P<0.01),SVF低、中、高浓度组压痛阈值与模型组相比均提高(P<0.01),且SVF高浓度组高于SVF低浓度组(P<0.05)。病理学结果表明,与正常组相比,模型组关节软骨破坏和退行性变化明显;SVF低、中、高浓度组相比于模型组关节软骨形态明显改善。模型组OARSI评分高于正常组(P<0.01),SVF低、中、高浓度组OARSI评分均较模型组下降(P<0.01)。结论SVF可提高KOA模型大鼠疼痛阈值,修复软骨损伤,且低剂量就有良好效果,是临床治疗KOA的潜在有效干预手段。 Objective To observe the effect of Stromal Vascular Fraction(SVF)on pain and cartilage renovation in rats with knee osteoarthritis(KOA).Methods SVF was prepared&om bilateral inguinal fat tissues of rats,and the pain models of KOA in rats were established by iodoacetic acid.25 rats were randomly divided into normal group,model group,SVF low-concentration group,SVF middle-concentration group,arid SVF high-concentration group,5 cases in each group.Using low(1.6×10^(4)个/mL),middle(3.2×10^(4)个/mL),and high(6.4×10^(4)个/mL)concentrations of SVF to treat bilateral knee joints in rats Injection treatment,once a week,and the rats'pain indicators were evaluated at the 2nd and 4th weeks after the model was replicated.Four weeks after the injection,the knee joints of each group of rats were taken for histopathological observation and OARSI score.Results At the 2nd week after the model was replicated,compared with the normal group,the tenderness threshold of the model group and the SVF low-,medium-,and high-concentration groups were reduced(P<0.01);at the 4th week after the model was replicated,the tenderness threshold of the model group lower than the normal group(P<0.01),the tenderness threshold of the SVF low-,medium-,and high-concentration groups were increased compared with the model group(P<0.01),and the SVF high-concentration group was higher than the SVF low-concentration group(P<0.05).The pathological results showed that compared with the normal group,the articular cartilage destruction and degeneration in the model group were obvious;the articular cartilage morphology of the SVF low-,medium-and high-concentration groups was significantly improved compared with the model group.The OARSI score of the model group was higher than that of the normal group(P<0.01),the OARSI score of the SVF low-,medium-,and high-concentration groups were all lower than that of the model group(P<0.01).Conclusion SVF can increase the pain threshold of KOA model rats,repair cartilage damage,and has a good effect at low doses.It is a potential effective intervention for cEnical treatment of KOA.
