摘要
目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)长链非编码RNA(lncRNA)ENST00000500949的表达变化及意义。方法选择2017年9月~2018年1月收治的SLE患者30例为研究对象。根据SLE疾病活动度(SLEDAI)评分将患者分为活动组18例(SLEDAI评分≥9分)、缓解组12例(SLEDAI评分<9分),同期选体检健康者20例作为健康对照组。Ficoll梯度密度原理分离纯化PBMC,采用生物芯片技术筛选SLE患者差异表达lncRNA,实时荧光定量法(qRT-PCR)检测PBMC中LncENST00000500949表达;魏氏法检测血沉(ESR),速率散射比浊法检测补体C3。Pearson线性相关分析指标间的相关性。结果 (1)本研究共筛选出163个差异表达lncRNA,其中表达上调基因118个,表达下调基因45个。选择表达差异较明显的lncENST00000500949作为观察指标。(2)qRT-PCR结果显示,与健康对照组相比,SLE活动组和缓解组PBMC中lncENST00000500949基因相对表达量分别为0.21±0.08,0.57±0.05,均明显降低,差异具有统计学意义(P均<0.05)。(3)进一步研究证实,活动组和缓解组ESR分别为64.28±10.25和30.75±9.43,均显著高于健康对照组(8.20±2.14,P均<0.05);活动组和缓解组补体C3含量分别为0.47±0.15和0.81±0.27,均显著低于健康对照组(1.35±0.26,P均<0.05);与缓解组比较,活动组ESR水平高、补体C3水平低(P均<0.05)。(4)Pearson相关分析结果显示,SLE患者PBMC中lncENST00000500949表达与ESR呈负相关关系(r=-0.523,P<0.05),与补体C3呈正相关(r=0.608,P<0.05)。结论 SLE患者PBMC中lncENST00000500949表达下调,其表达变化在一定程度上可反映疾病活动度,可作为评估SLE疾病活动度的潜在生物标志物。
Objective To investigate the role and clinical significance of Long noncoding RNA(lncRNA) ENST00000500949 in the progression of systemic lupus erythematosus(SLE).Methods Thirty SLE patients admitted from September 2017 to January 2018 were selected as research objects.The expression of peripheral blood mononuclear cells(PBMCs) in 30 patients with SLE(18 in the active group and 12 in the remission group) and 20 healthy controls was verified by qRT-PCR(quantitative Real-time RT-PCR) method for differential expression of lncRNA ENST00000500949.At the same time, the relationship between lncRNA ENST00000500949 and SLE clinical activity indicators was analyzed.Results(1)In this study, a total of 163 significant differentially expressed lncRNAs were screened by chip technology, of which 118 were up-regulated and 45 were down-regulated.(2)Results of qRT-PCR showed that the relative expression level of lncENST0000500949 gene in SLE activity group and remission group was 0.21±0.08 and 0.57±0.05 respectively, which was decreased significantly compared with health control group(P<0.05).(3)ESR of SLE activity group was significantly higher than SLE remission group(P<0.05) and healthy control group(P<0.05).The complement C3 of SLE activity group was significantly lower than that of SLE remission group(P<0.05) and healthy control group(P<0.05).Compared with healthy control group, SLE remission group had higher ESR expression and decreased expression of complement C3(P<0.05).(4)Pearson correlation analysis: The expression of lncRNA ENST00000500949 in SLE patients was negatively correlated with ESR(r=-0.523,P<0.05)and negatively positively correlated with complement C3(r=0.608,P<0.05).Conclusion The expression of lncRNA ENST00000500949 in PBMC of SLE patients is down-regulated, and its expression changes can reflect disease activity to a certain extent, which can be used as a potential biomarkers to evaluate disease activity in SLE.
