甲状腺乳头状癌患者外周血miR-146a、miR-146b表达水平与预后的关系研究

Expressions of Serum MiR-146a and MiR-146b in Papillary Thyroid Carcinoma and Their Relationships with Clinicopathological Features and Prognosis

目的探讨甲状腺乳头状癌(papillary thyroid cancer,PTC)患者血清miR-146a、miR-146b的表达水平及其与临床病理特征、预后的关系。方法采用前瞻性研究方法,选取2018年6月至2020年2月本院收治的PTC患者80例为研究对象,50例同期健康人群作为对照组。荧光定量PCR检测外周血中miR-146a、miR-146b的表达水平,分析PTC患者血清miR-146a、miR-146b水平与临床病理、预后的关系。结果PTC患者血清miR-146a、miR-146b表达水平(5.18±0.04、2.59±0.13)高于正常人群(1.01±0.01、1.12±0.03)(t=23.091、8.431,P<0.001、<0.001)。miR-146a、miR-146b表达与TNM分期、分化程度、淋巴结转移和组织学亚型有显著相关性(P<0.05)。miR-146a低表达组患者累积生存率高于miR-146a高表达组(P=0.001);miR-146b低表达组患者累积生存率高于miR-146b高表达组(P<0.001)。结论miR-146a、miR-146b在PTC患者血清中高水平表达,与肿瘤的进展、预后不良有关。

Objective To investigate the expression levels of serum miR-146a and miR-146b in patients with papillary thyroid cancer(PTC)and their relationships with clinicopathological features and prognosis.Methods 80 patients with PTC from June,2018 to February,2020 in our hospital were selected as the research objects,while 50 healthy people during the same period were selected as the control group.The expression levels of miR-146a and miR-146b in peripheral blood were detected by fluorescence quantitative PCR,and the relationships between serum levels of miR-146a and miR-146b and clinicopathological features and prognosis were analyzed.Results The expression levels of miR-146a and miR-146b in patients with HCC(5.18±0.04,2.59±0.13)were higher than those in normal people(1.01±0.01,1.12±0.03)(t=23.091,8.431,P<0.001,<0.001).The expressions of miR-30a and mir-106b were significantly correlated with TNM stages,differentiation degrees,lymph node metastasis and histological subtypes(P<0.05).The cumulative survival rate of patients with low expression of miR-146a was higher than that of patients with high expression of miR-146a(P=0.001),while the cumulative survival rate of patients with low expression of miR-146b was higher than that of patients with high expression of miR-146b(P<0.001).Conclusion High expressions of miR-146a and miR-146b in serum of patients with PTC are related to tumor progression and poor prognosis.
